【摘要】：為了研究奶牛黃體的形成及退化過程的相關機理,并進一步探究更加準確的妊娠及相關生理過程的蛋白標記物,試驗采用蛋白質組學經典的二維凝膠電泳(2-DE)技術檢測和分析妊娠前后黃體組織間的差異蛋白,使用PDQuest8.0軟件檢測,取差異蛋白點進行分析,并經MALDI TOF/TOFMS/MS檢測。結果表明:6種差異蛋白分別為26s蛋白酶體蛋白、纖維蛋白、DNA拓撲異構酶蛋白、核纖層蛋白、硫氧還原蛋白、微管蛋白。對這6種蛋白在組織間的表達差異進行分析,發(fā)現(xiàn)這些蛋白可能與黃體形成和黃體退化及黃體激素分泌有關。
[Abstract]:In order to study the mechanism of luteal formation and degeneration in dairy cows, and to explore more accurate protein markers of pregnancy and related physiological processes, Two dimensional gel electrophoresis (2-DE), a classical proteomics technique, was used to detect and analyze differentially expressed proteins in corpus luteum before and after pregnancy. PDQuest8.0 software was used to detect the differential proteins and MALDI TOF/TOFMS/MS was used to detect them. The results showed that the six differential proteins were 26s proteasome protein, fibrin DNA topoisomerase protein, nuclear laminar protein, thioxy-reducing protein and tubulin. It was found that these proteins might be related to luteal formation, luteal degeneration and luteal hormone secretion.
