【摘要】：為探究Kiss-1和GPR54基因在山羊季節(jié)性繁殖中的作用,分別在四川省金堂縣和理縣各選取5只10月齡處于發(fā)情前期的藏山羊和金堂黑山羊為研究對象,對Kiss-1和GPR54基因編碼區(qū)進行序列及組織表達分析。結(jié)果顯示:金堂黑山羊和藏山羊Kiss-1基因編碼區(qū)均長408 bp,編碼135個氨基酸,兩品種間存在2處同義突變。GPR54基因編碼區(qū)均長244 bp,編碼81個氨基酸,兩品種間存在1處同義突變;金堂黑山羊Kiss-1和GPR54基因CDs區(qū)核苷酸序列與藏山羊的同源性分別為99.5%、99.6%;Kiss-1和GPR54基因在兩品種山羊的卵巢、子宮、輸卵管、垂體中均有表達,但只有Kiss-1基因在金堂黑山羊垂體中的表達量顯著高于藏山羊(P0.05),而在其他組織品種間差異不顯著(P0.05)。提示:Kiss-1基因可能參與調(diào)控山羊季節(jié)性繁殖,而GPR54基因是否是引起季節(jié)性繁殖的原因還有待進一步研究。
[Abstract]:In order to investigate the role of Kiss-1 and GPR54 genes in seasonal reproduction of goats, five Tibetan goats and Jintang black goats, aged 10 months, were selected from Jintang County and Lixian County, Sichuan Province, respectively. The sequence and tissue expression of Kiss-1 and GPR54 gene coding region were analyzed. The results showed that the coding region of Kiss-1 gene in Jintang Black Goat and Tibetan Goat was 408bplong, encoding 135 amino acids. There were 2 synonymous mutations. The coding region of GPR54 gene was 244bp, encoding 81 amino acids, and there was one synonymous mutation between the two breeds. The nucleotide sequence of CDs region of Kiss-1 and GPR54 gene in Jintang Black Goat was 99.55.The nucleotide sequence of Kiss-1 and GPR54 genes were 99.5% and 99.6% respectively in ovary, uterus, fallopian tube and pituitary gland of the two breeds of goats. But only the expression of Kiss-1 gene in pituitary gland of Jintang black goat was significantly higher than that of Tibetan goat (P0.05), but there was no significant difference among other tissue varieties (P0.05). It is suggested that the gene of: Kiss-1 may be involved in the regulation of seasonal reproduction in goats, and whether the GPR54 gene is the cause of seasonal reproduction remains to be further studied.
