百里香酚對山羊子宮內(nèi)膜上皮細(xì)胞的體外毒活性研究

【摘要】:為了探究百里香酚對山羊子宮內(nèi)膜上皮細(xì)胞(goat endometrial epithelial cells,g EECs)的毒性作用,采用不同濃度的百里香酚作用于g EECs后,觀測其對細(xì)胞生長曲線,細(xì)胞形態(tài)學(xué)變化,以及細(xì)胞膜結(jié)構(gòu)完整性的影響。結(jié)果表明,百里香酚對EECs的毒性作用與其濃度呈劑量依賴效應(yīng)(y=-0.008x+1.2202,R~2=0.9767),即隨著藥物濃度的增加細(xì)胞的增殖抑制率增大,藥物對細(xì)胞的半數(shù)增殖抑制率(IC50)為100μg/m L。當(dāng)50、100μg/m L的百里香酚作用于EECs時(shí),g EECs的細(xì)胞核、細(xì)胞漿的形態(tài)均未產(chǎn)生明顯變化,但當(dāng)150μg/m L的百里香酚作用于g EECs時(shí),細(xì)胞膜破裂,細(xì)胞死亡;而且藥物濃度≥100μg/m L時(shí)細(xì)胞LDH的釋放量顯著高于正常細(xì)胞組,g EECs細(xì)胞膜的完整性受到破壞。百里香酚作用于山羊子宮內(nèi)膜上皮細(xì)胞后呈現(xiàn)一定的毒性作用,且毒性作用和藥物的濃度有關(guān)。百里香酚作用于g EECs的安全范圍在0~100μg/m L,研究結(jié)果為百里香酚治療奶牛等經(jīng)濟(jì)動(dòng)物的炎癥性疾病的臨床用藥濃度提供理論依據(jù)。
