豬繁殖與呼吸綜合征病毒RT-LAMP快速檢測(cè)方法的建立

【摘要】:為建立一種快速、靈敏、簡便的豬繁殖與呼吸綜合征病毒(PRRSV)早期檢測(cè)方法,本研究利用逆轉(zhuǎn)錄環(huán)介導(dǎo)恒溫?cái)U(kuò)增技術(shù)(RT-LAMP),針對(duì)PRRSV的ORF5基因片段設(shè)計(jì)了4條引物,利用Bst DNA聚合酶在65℃恒溫條件下進(jìn)行逆轉(zhuǎn)錄擴(kuò)增,通過1.0%瓊脂糖凝膠電泳和加入SYBR GreenⅠ染料肉眼判斷結(jié)果,建立了PRRSV RT-LAMP檢測(cè)方法。結(jié)果表明,該檢測(cè)方法具有良好的特異性,與其他常見病毒如豬瘟病毒、豬圓環(huán)病毒2型、豬細(xì)小病毒、豬偽狂犬病病毒等無交叉反應(yīng),較普通RT-PCR靈敏性高100倍。采用RT-LAMP和RT-PCR分別對(duì)10份臨床樣本同時(shí)進(jìn)行檢測(cè),符合率為100%。因此,本研究建立的RT-LAMP是一種可適用于臨床PRRSV檢測(cè)的快速、簡單、靈敏、特異的檢測(cè)方法。
