【摘要】：建立了一種測(cè)定比格(Beagle)犬尿液中檸檬酸濃度的方法,利用檸檬酸與溴化鉀、高錳酸鉀在酸性溶液中生成五溴丙酮,五溴丙酮經(jīng)石油醚(90℃~120℃)萃取后,與硫脲反應(yīng)生成水溶性黃色復(fù)合物,其用紫外分光光度計(jì)在435nm波長處測(cè)定。結(jié)果顯示,檸檬酸濃度在0.005mg/mL~0.25mg/mL范圍內(nèi),水溶性黃色復(fù)合物的吸光度與檸檬酸的濃度呈良好的線性關(guān)系,OD435=1.8335C+0.0038,r=0.9996,平均回收率為99.954%(n=5),變異系數(shù)為1.64%(n=20)。該方法操作簡(jiǎn)單,快速準(zhǔn)確,適用于非臨床研究中Beagle犬尿液中檸檬酸濃度的測(cè)定。
