奶牛子宮內(nèi)膜炎抗菌蒙藥復(fù)方的篩選及其抗菌作用的研究

【摘要】:奶牛子宮內(nèi)膜炎是引發(fā)奶牛不孕癥的主要原因之一,因其較高的發(fā)病率,使得我國(guó)的奶牛產(chǎn)業(yè)遭受重大的經(jīng)濟(jì)損失。奶牛子宮內(nèi)膜炎最為常用的治療方法是抗生素和其他化學(xué)藥物子宮內(nèi)灌注和子宮內(nèi)沖洗。但是,由濫用抗生素和其他化學(xué)藥物引起的抗生素殘留和耐藥菌株產(chǎn)生數(shù)量及速度的不斷增加,需求一種能夠治療奶牛子宮內(nèi)膜炎的綠色強(qiáng)效經(jīng)濟(jì)藥物成為一個(gè)亟待解決的問(wèn)題。蒙藥具有抗菌、抗炎免疫和寒熱調(diào)理作用,通過(guò)現(xiàn)代的研究方法,希望能從中篩選出有效治療奶牛子宮內(nèi)膜炎的藥物組方。主要研究?jī)?nèi)容及結(jié)果如下:1、以常規(guī)方法從奶牛子宮內(nèi)膜炎分泌物中分離純化主要致病菌,經(jīng)分離純化以及生化鑒定所得主要致病菌為大腸桿菌和金黃色葡萄球菌,其中大腸桿菌64株,金黃色葡萄球菌17株。2、以玻片凝集法對(duì)分離所得的64株大腸桿菌進(jìn)行抗“O”血清型分析,鑒定結(jié)果為33株11種血清型,即O86、O1、O126、O111、O29、O28、O8、O91、O2、O26、O25。其中以0s6鑒定率最高為14%,為優(yōu)勢(shì)血清型,其余菌株有4株出現(xiàn)自凝集現(xiàn)象,27株未能檢測(cè)出。3、對(duì)大黃、連翹、訶子等26味蒙藥經(jīng)體外抑菌試驗(yàn)結(jié)合正交方法,篩選蒙藥復(fù)方并對(duì)其進(jìn)行劑量?jī)?yōu)化,得到4組蒙藥復(fù)方,分別為復(fù)方Ⅰ:大黃70g、丹參50g;復(fù)方Ⅱ:黃芩70g、大黃50g;復(fù)方Ⅲ:大黃、訶子各50g,連翹30g;復(fù)方Ⅴ:黃芩、大黃、訶子各70g。4、以昆明系小鼠為試驗(yàn)動(dòng)物,測(cè)定篩選出的復(fù)方對(duì)小鼠(攻毒菌量為80%混合菌致死量)的保護(hù)率。結(jié)果為復(fù)方Ⅰ、Ⅱ、Ⅲ、Ⅴ對(duì)攻毒小鼠的保護(hù)率分別為40%、40%、60%和50%,與環(huán)丙沙星組對(duì)比,其中復(fù)方Ⅴ與環(huán)丙沙星組效果相當(dāng),復(fù)方Ⅲ高于環(huán)丙沙星組。5、以體外抑菌方法測(cè)定蒙藥復(fù)方的抗菌譜,結(jié)果為無(wú)乳鏈球菌、鼠傷寒沙門氏菌、腸出血性大腸桿菌、金黃色葡萄球菌、銅綠假單胞菌、蠟樣芽孢桿菌對(duì)4組復(fù)方均為高敏感或極敏感,其中復(fù)方Ⅱ的抑菌圈直徑顯著高于其他復(fù)方(P0.05)。6、以酶標(biāo)比濁法測(cè)定復(fù)方藥物對(duì)兩種致病菌生長(zhǎng)曲線的影響,微生物粘附法測(cè)定復(fù)方藥物對(duì)菌體細(xì)胞表面疏水性的影響,鄰硝基苯-β-D-半乳吡喃糖苷(ONPG)法測(cè)定復(fù)方藥物對(duì)菌體細(xì)胞膜滲透性的影響,測(cè)定堿性磷酸酶(APK)活性法測(cè)定復(fù)方藥物對(duì)菌體細(xì)胞壁的影響。所得結(jié)果為4組蒙藥復(fù)方均能很好地影響致病菌的生長(zhǎng)曲線,且都能夠從不同程度上影響菌體細(xì)胞表面的疏水性以及其細(xì)胞膜的滲透性,對(duì)菌體細(xì)胞壁也有不同程度的損傷。
[Abstract]:Endometritis is one of the main causes of infertility in dairy cows. Because of its high incidence, the dairy industry in China has suffered great economic losses. The most commonly used treatment for endometritis in cows is intrauterine perfusion and irrigation with antibiotics and other chemicals. However, the number and rate of production of antibiotic residues and resistant strains resulting from the abuse of antibiotics and other chemical drugs is increasing, The demand for a green economic drug for the treatment of dairy endometritis has become an urgent problem. Mongolian medicine has antimicrobial, anti-inflammatory, immune and cold-heat conditioning effects. Through modern research methods, it is hoped that it can be used to screen effective prescriptions for the treatment of endometritis in dairy cows. The main research contents and results are as follows: 1. The main pathogenic bacteria were isolated and purified from the secretion of endometritis of dairy cows by routine method. The main pathogenic bacteria were Escherichia coli and Staphylococcus aureus after isolation, purification and biochemical identification. Among them, 64 strains of Escherichia coli and 17 strains of Staphylococcus aureus were analyzed by slide agglutination method. The results showed that 33 strains of 11 serotypes, namely O86O1O126O111O29O28O28O91O2O26O25. Among them, the 0s6 identification rate was the highest, and the dominant serotype was 14. The other 4 strains showed self-agglutination phenomenon. 27 strains could not be detected. 26 Mongolian drugs, such as rhubarb, Forsythia suspensa and Fructus chebula, were tested by in vitro bacteriostasis test and orthogonal method.
