川金果欖油膏治療大鼠燙傷的藥效學(xué)觀察
[Abstract]:Burn is a disease of tissue damage caused by various physical or chemical factors, including hydrothermal, steam, flame, electric energy, chemical material, radiating line and so on. The study of Chinese medicine treatment for frequent burns is also in depth. This subject is mainly to observe the protective effect of the ointment on scald wounds in rats and further explore its effect. Mechanism to provide a solid theoretical basis for promoting the clinical application of Chuanjin olive oil ointment and the development of new dosage forms. The main experimental contents and results are as follows: 1 rats with shallow second degree scald model of 1 rats were established by boiling water heated 100g weights to 5S, 10s, 15s and 20s respectively, and scalded after 24h were cut to scald after scalding 24h. All layers of skin, pathological sections were prepared, and the pathological changes of skin epidermis and dermis were observed. Conclusion: scald 5S can establish a stable II degree scald model in rats, scald 10s, 15s, 20s can establish different degree of rat deep II scald model.2 Sichuan golden fruit olivan ointment to protect the scald wound of rats, SD rats 20, male and female. Half, randomly divided into 5 groups, each group was 4. After the injury, 30min was given to the scald parts of each group respectively. The wound was evenly covered with sterilized gauze, 1 times a day, the drug was given to the wound to complete healing. The model group was treated with ointment matrix. The complete cover was the basis of the complete healing of the wound. Compared with the model group, the high dose group (P0. 01) and medium dose group (P0.05) could significantly shorten the healing time of the wound. Compared with the model group, the healing rate of the high dose group and the medium dose group increased significantly after the injury (P0.05), and the 14d also continued the trend after the injury. From the pathological histological sections of the scalded rats during the whole healing process, the Sichuan golden ointment could reduce the early inflammation. The infiltration of the disease cells, quickening the growth of fibroblast and capillary in the wound, accelerating the skin cover in the later period, promoting the healing of the wound healing.3 and promoting the healing of scald wound in rats, 84 rats and male and male 4 rats were divided into 5 groups randomly, and the other rats were randomly divided into 5 groups, 16 rats in each group. After scald, the groups were given the drugs respectively. In the treatment, 4 rats were randomly selected from 1D, 3D, 7d, and 14d after injury. The blood samples from the tail vein were collected from the blood serum from the tail vein. The tissue specimens were cut from the wound tissue and stored at -80 fridge. Through the measurement of some physiological indexes of 1d-14d after scald in experimental rats, it was found that the increasing trend of IL-6 content in the serum of scalded rats was obvious and reached the third day after injury. The peak value was significantly different from that of the normal group (P0.01), which indirectly reflected the success of the mold making. From the content of SOD and MDA in the wound tissue, the burn wound had a continuous deepening process in the early stage. The content of hydroxyproline in the wound surface of the drug group were all in the third Tianda peak in the post injury, earlier than the model group, and the peak concentration was significantly higher than that in the model group. Seventh Compared with the model group, the water content of the wound tissue in the low dose group of the Sichuan golden olive oil ointment decreased significantly (P0.05), and the effect was more obvious (P0.01) fourteenth days after injury. The activity of SOD in the medium and low dose group was very significant (P0.01), and the content of MDA in each group also kept declining, and the decrease of the high dose group was large, and the model was large. The peak time of EGF content in the wound tissue of each drug group was earlier than that in the model group, and the content of EGF in the wound tissue in the middle dose group was higher than that in the model group (P0.05) in the middle dose group seventh days after the injury, and the IL-6 content in the serum of the scalded rats decreased with the time prolonging, which was compared with the model group. The decreasing trend of the high dose group (P0.01) and the medium dose group (P0.05) was more obvious, and the content of TGF-p in the serum of each group was seventh Tianda peak after injury, and compared with the model group, the content of TGF-p in the serum of scalded rats increased significantly (P0.01). Conclusion: 1. using the 100g weight of 5 minutes heating for 5 minutes to induce the injury of 5S can establish a stable shallow rat. The II degree scald model.2. Sichuan golden ointment ointment can improve the healing rate of burn wound at different time points, shorten the healing time of scald wound, and show a better repair effect on scald wound. The mechanism of.3. Kaga ruga ointment to promote the healing of scald wound may involve the formation of collagen in the wound surface and the inhibition of lipid peroxidation. It can promote cell proliferation and epidermal crawling, control excessive synthesis and release of some inflammatory mediators, and promote the increase of EGF and TGF-p contents in scalded rats.
