【摘要】：為獲得陜西某羊場病羊膿包中的病原菌,以無菌采集的病羊頸部膿汁為材料,進行細菌分離培養(yǎng),鑒定分離菌株的形態(tài)特征、培養(yǎng)特性、生化特性,挑取分離菌株進行協(xié)同溶血試驗(cAMP)和16SrRNA基因的PCR擴增,并對序列進行分析,NJ法構(gòu)建系統(tǒng)發(fā)育樹;用分離菌株分別接種Balb/c小鼠和奶山羊;藥敏試驗鑒定分離株的耐藥性。結(jié)果顯示,分離菌株的形態(tài)特征、培養(yǎng)特性、生化特性與伯杰細菌鑒定手冊所述偽結(jié)核棒狀桿菌特性一致;測序顯示分離菌株與GenBank中已收錄偽結(jié)核棒狀桿菌相似性達97%以上;Balb/c小鼠致病性試驗和奶山羊動物回歸試驗表明分離株有強致病性;藥敏試驗表明分離菌株對紅霉素、環(huán)丙沙星、克拉霉素、強力霉素、四環(huán)素、頭孢唑啉、新霉素、氧氟沙星等多種抗生素敏感,對復(fù)方新諾明耐藥。
[Abstract]:In order to obtain the pathogenic bacteria in the abscess of sick sheep in a sheep farm in Shaanxi Province, the bacteria were isolated and cultured in the neck pus collected from the sterile sheep, and the morphological characteristics, culture characteristics and biochemical characteristics of the isolated strains were identified. The isolated strains were selected for co-hemolysis test (camp) and PCR amplification of 16s rRNA gene. The phylogenetic tree was constructed by NJ method. The isolated strains were inoculated with Balb / c mice and dairy goats respectively. The drug sensitivity test was used to identify the drug resistance of the isolated strains. The results showed that the morphological characteristics, culture characteristics and biochemical characteristics of the isolated strains were consistent with those of Corynebacterium tuberculosis described in Berger's Manual of bacterial Identification. The results of sequencing showed that the similarity between the isolated strains and pseudotuberculosis Corynebacterium tuberculosis was more than 97% in Balb / c mice and in dairy goat animal regression tests, and the drug sensitivity test showed that the isolates had strong pathogenicity to erythromycin and ciprofloxacin. Clarithromycin, doxycycline, tetracycline, cefazolin, neomycin, ofloxacin and other antibiotics are sensitive and resistant to compound sulfamide.
