妊娠后期宮內生長限制對蒙古綿羊胎兒胸腺細胞增殖影響及機理研究

【摘要】:本論文系統(tǒng)的闡述了妊娠后期IUGR對蒙古綿羊胎兒胸腺細胞增殖的相關因子表達的影響。選取24只經同期妊娠的健壯蒙古綿羊作為研究對象。妊娠期90 d時,挑取6只重量相近的母羊進行屠宰,余下18只綿羊根據重量隨機分為3個不同營養(yǎng)水平處理組:0.18 MJME-BW-0.75·d-1 (RG1:rrn=6),0.33 MJME·BW0-0.75·d-1(RG2:n=6)和自由采食(CG:n=6),將剩余三個營養(yǎng)水平飼喂至妊娠期140 d,剩余各組6只妊娠羊進行屠宰試驗。試驗結果表明:(1)妊娠后期IUGR引起RG1組胎兒體重(p0.01)、胸腺重(p0.01)、總DNA含量(p0.01)、細胞增殖指數(p0.05)、PCNA個數(p0.01)極顯著低于CG組,G0/G1期細胞比例明顯高于CG組(p0.05)。RG2組胎兒體重(p0.01)胸腺重(p0.05)、總DNA含量(p0.05)顯著低于CG組,IUGR胎兒胸腺的DNA合成受限制程度顯著,是導致細胞增殖受限的主要原因之一。(2)RG1組GHR (p0.01)、IGF-1R (p0.01)、IGF-2R (p0.01)、CyclinB1 (p0.05)、CyclinD (p0.05)、CDK1 (p0.05)、CDK2 (p0.01)、CDK4 (p0.05)、 E2F1 (p0.01)、E2F4 (p0.01)、E2F5 (p0.01)基因表達量顯著低于CG組,RG2組GHR (p0.01)、IGF-1R (p0.05)、CDK1 (p0.05)、CDK2 (p0.01)、CDK4 (p0.01)、E2F1 (p0.05)、E2F5 (p0.01)基因表達均顯著低于CG組,IGF-2R、 CyclinB1、E2F4呈降低趨勢,但差異均不顯著(p0.05)。(3)RG1組GH (p0.01)、IGF-1 (p0.05)、IGF-2 (p0.01)、GHR(p0.05) IGF-1R (p0.05)、IGF-2R (p0.05)、CyclinA (p0.05)、CyclinB (p0.05) CyclinD (p0.05)、CyclinE (p0.05)、E2F2 (p0.05)均顯著低于CG組,IGFBP-1顯著高于CG組(p0.05),E2F1、E2F4、E2F5均呈增高趨勢,但差異不顯著(p0.05),CDK1、CDK2、CDK4呈降低趨勢,但差異均不顯著(p0.05);RG2組IGFBP-2 (p0.01)、GHR (p0.05)顯著低于CG組,IGFBP-1顯著高于CG組(p0.05)。CyclinA、CyclinB、CyclinD、CyclinE、CDK1、CDK2、CDK4、E2F2蛋白水平表達呈降低趨勢,但差異不顯著(p0.05), E2F1、E2F4、E2F5均呈增高趨勢,但差異均不顯著(p0.05)。
[Abstract]:The effects of IUGR on the expression of factors related to the proliferation of fetal thymocytes in Mongolian sheep were systematically discussed in this paper. Twenty-four healthy Mongolian sheep with simultaneous pregnancy were selected as study objects. At 90 days gestation, 6 ewes of similar weight were selected and slaughtered. The remaining 18 sheep were randomly divided into three groups: 0.18 MJME-BW-0.75 d-1 (RG1: rrrnnf6) 0.33 MJME BW0-0.75 d-1 (RG2: nrnf6) and CG2: nong6 according to their weight. The remaining three nutritional levels were fed to 140 days of gestation, and six sheep in each group were slaughtered. The results showed that: (1) the fetal weight (p0.01), thymus weight (p0.01), total DNA content (p0.01), proliferative index (p0.05) PCNA number (p0.01) of IUGR induced by IUGR in RG1 group were significantly lower than those in CG group (p0.05) 路RG2 group (p0.01).
