益生菌對(duì)雛雞免疫器官免疫功能及β-防御素-1mRNA表達(dá)的影響

【摘要】:以1日齡雛雞為研究對(duì)象,應(yīng)用熒光定量PCR等技術(shù),結(jié)合器官組織石蠟切片及組織化學(xué)染色等方法,通過免疫器官——胸腺、脾臟、法氏囊雞β-防御素-1(Gal-1)mRNA表達(dá),漿細(xì)胞數(shù)量及其核漿比,TANAE+細(xì)胞數(shù)量等有關(guān)指標(biāo)動(dòng)態(tài)變化的檢測(cè),較全面系統(tǒng)的研究了不同種類益生菌對(duì)雛雞免疫器官免疫功能和Gal-1mRNA表達(dá)的影響,旨在為益生菌及其制劑在動(dòng)物生產(chǎn)中的應(yīng)用及其研發(fā)新產(chǎn)品提供科學(xué)實(shí)驗(yàn)依據(jù),研究結(jié)果發(fā)現(xiàn):(1)雛雞單獨(dú)應(yīng)用雙歧桿菌后,胸腺、脾臟、法氏囊中TANAE+細(xì)胞數(shù)量均于7~13日齡時(shí)較對(duì)照雛雞明顯增多(P0.05);脾臟、法氏囊和胸腺中的漿細(xì)胞數(shù)量分別于10~13日齡和7~10日齡時(shí)較對(duì)照雛雞明顯增多(P0.05);但Gal-1mRNA表達(dá)于10~13日齡時(shí)較對(duì)照雛雞明顯提高(P0.05或P0.01)。(2)雛雞單獨(dú)應(yīng)用乳酸桿菌后,脾臟和法氏囊中漿細(xì)胞和TANAE+細(xì)胞數(shù)量以及Gal-1m RNA均于7~13日齡時(shí)顯著或極顯著高于對(duì)照組雛雞(P0.05或P0.01),而胸腺中漿細(xì)胞和TANAE+細(xì)胞數(shù)量于7~10日齡時(shí)明顯多于對(duì)照組雛雞(P0.05),但Gal-1mRNA表達(dá)于7日齡時(shí)較對(duì)照雛雞明顯提高(P0.05或P0.01)。(3)雙歧桿菌和乳酸桿菌聯(lián)合應(yīng)用雛雞后,胸腺、脾臟和法氏囊中的TANAE+細(xì)胞數(shù)量分別于7~10、13和10~13日齡時(shí)明顯多于對(duì)照組雛雞(P0.05);免疫器官中的漿細(xì)胞均與7日齡時(shí)明顯多于對(duì)照組雛雞(P0.05);但Gal-1mRNA表達(dá)于10~13日齡時(shí)較對(duì)照雛雞明顯提高(P0.05或P0.01)。乳酸桿菌對(duì)雛雞免疫器官T淋巴細(xì)胞和漿細(xì)胞數(shù)量及Gal-1m RNA表達(dá)的作用效果優(yōu)于雙歧桿菌。
[Abstract]:The expression of 尾 -defensin-1 (Gal-1) mRNA in the immune organs of thymus, spleen and Fabricius chicks was studied by fluorescence quantitative PCR, paraffin section and histochemical staining. The effects of different probiotics on immune function of immune organs and expression of Gal-1 mRNA in chicks were studied comprehensively and systematically by detecting the dynamic changes of plasma cell number and the number of TANAE cells, such as the number of plasma cells and the number of TANAE cells. The aim of this study was to provide scientific experimental basis for the application of probiotics and their preparations in animal production and to develop new products. The results showed that: (1) the thymus and spleen of chicks were treated with bifidobacterium alone. The number of TANAE cells in the bursa of Fabricius was significantly higher than that of the control chicks at the age of 7 ~ 13 days (P0.05). The number of plasma cells in the bursa of Fabricius and thymus was significantly higher than that in the control group at the age of 10 and 10 days (P0.05), but the expression of Gal-1 mRNA in the thymus was significantly higher than that in the control (P0.05 or P0.01). However, the expression of Gal-1 mRNA at 7 days old was significantly higher than that in the control (P0.05 or P0.01). (3) after the combination of Bifidobacterium and Lactobacillus. Thymus gland, The number of TANAE cells in spleen and bursa of Fabricius was significantly higher than that in the control group at the age of 7 days (P0.05), the plasma cells in the immune organs were significantly higher than those in the control group (P0.05), but the expression of Gal-1 mRNA in the spleen and bursa of Fabricius was significantly higher than that in the control group at the age of 1013 days (P0.05), but the expression of Gal-1 mRNA was higher than that in the control group at the age of 1013 days. Chicken significantly increased (P0.05 or P0.01). The effect of Lactobacillus on the number of T lymphocytes and plasma cells and the expression of Gal-1m RNA in chicken immune organs was better than that of Bifidobacterium.
