利用細(xì)菌性腦膜炎鼠模型評價(jià)側(cè)腦室給藥的治療效果

【摘要】:側(cè)腦室給藥技術(shù)是一種新型的給藥技術(shù),不僅可以用于大腦中樞疾病的治療,還廣泛用于研究大腦心室的形成和形態(tài)發(fā)生以及描述大腦形態(tài)發(fā)生的突變體,并且側(cè)腦室注射給藥能直接將藥物作用于大腦中樞系統(tǒng),從而更有效的發(fā)揮了其治療或損傷作用,在臨床實(shí)驗(yàn)中具有較大的意義。目的:建立傳染性胸膜放線桿菌引起的細(xì)菌性腦膜炎鼠模型,比較側(cè)腦室和靜脈注射連續(xù)3天給于青霉素對大鼠腦膜炎模型的治療效果。實(shí)驗(yàn)方法:實(shí)驗(yàn)需使用16只雄性的成年SD大鼠且本次實(shí)驗(yàn)動物體重范圍為250-300g,將其任意分成4個(gè)組,實(shí)驗(yàn)前2天選取其中一組進(jìn)行腦部導(dǎo)管植入手術(shù)。實(shí)驗(yàn)時(shí)通過異氟烷將動物麻醉,用微量注射器作小腦延髓池穿刺,接種胸膜肺炎放線菌菌懸液,建立胸膜肺炎放線菌感染鼠的腦膜炎模型,給予不同給藥劑量進(jìn)行為期3天治療,經(jīng)側(cè)腦室與靜脈多次注射青霉素鈉,通過對實(shí)驗(yàn)大鼠的存活率、臨床癥狀的觀察,腦病理組織學(xué)檢查、細(xì)菌培養(yǎng)及聚合酶鏈反應(yīng)(PCR)等技術(shù)對不同給藥途徑的效果評估。結(jié)果:大鼠接種傳染性胸膜肺炎放線桿菌后,均出現(xiàn)腦膜炎臨床癥狀;PCR實(shí)驗(yàn)檢測結(jié)果呈陽性,證明腦脊液中檢測到APP 7型菌株。大鼠細(xì)菌性腦膜炎模型制備成功。給藥組大鼠臨床癥狀較模型對照組有明顯改善;側(cè)腦室給予0.01mpk治療效果要優(yōu)于靜脈注射10mpk,跟靜脈注射62.5mpk組治療效果大體相當(dāng)。結(jié)論:側(cè)腦室給藥給藥劑量低,治療效果好,是一種理想的細(xì)菌性腦膜炎治療方法。
[Abstract]:Lateral ventricle administration is a new kind of drug delivery technology. It can be used not only in the treatment of brain diseases, but also in the study of the formation and morphogenesis of the ventricles of the brain and the mutants describing the morphogenesis of the brain. The injection of the lateral ventricle can directly effect the drug on the central brain system, thus making it more effective. The effect of its treatment or injury is of great significance in clinical trials. Objective: to establish a bacterial meningitis rat model caused by contagious Actinobacillus pleurobacilli and compare the therapeutic effect of penicillin to the meningitis model of rats by penicillin for 3 days in the lateral ventricle and intravenous injection. Experimental methods should be used in 16 adult male SD rats. The weight range of the second experimental animal was 250-300g, which was randomly divided into 4 groups, and one of them was implanted in the brain 2 days before the experiment. In the experiment, the animals were anesthetized with isoflurane, the micro syringe was used as the cerebellum medullary pool, and the actinomycete suspension was inoculated to establish the meningitis model of the actinomycete infected mice. Type, given the dosage of different dosage for 3 days of treatment, through the lateral ventricle and intravenous injection of penicillin sodium, through the experimental rats, the survival rate, clinical symptoms, brain histopathology examination, bacterial culture and polymerase chain reaction (PCR) technique to evaluate the effect of different drug delivery methods. Results: the rats were inoculated with infectious pleural lung. The clinical symptoms of meningitis were found after Actinobacillus inflammatory. The results of PCR test were positive. It was proved that APP 7 was detected in the cerebrospinal fluid. The model of bacterial meningitis in rats was successfully prepared. The clinical symptoms of the rats in the administration group were obviously better than those in the model control group; the effect of 0.01mpk in the lateral ventricle was better than the intravenous injection of 10mpk and vein. Conclusion: the dosage of the lateral ventricle is low and the therapeutic effect is good. It is an ideal treatment method for bacterial meningitis. 62.5mpk is a safe and effective drug for the treatment of bacterial meningitis.
