犬弓首蛔蟲線粒體cox1基因測序鑒定及生物學(xué)特性研究

【摘要】:犬弓首蛔蟲屬于蛔目(Ascarididea)弓首科(Toxocaridae)弓首屬(Toxocara),寄生于犬和其它動物(包括人)的小腸和胃內(nèi)的一種線蟲。本研究為了了解犬弓首蛔蟲蟲卵在體外發(fā)育情況、鑒別其感染性蟲卵,掌握幼蟲在小雞體內(nèi)移行情況,而做了一系列的相關(guān)研究,為犬弓首蛔蟲病的診斷和防治提供了大量科學(xué)依據(jù)。本研究的第一部分對7株犬蛔蟲cox1基因進(jìn)行測序與序列分析。結(jié)果顯示7株犬蛔蟲cox1基因序列長度為440bp。線粒體cox1基因的種內(nèi)相似性為96.7%~100%,種間相似性為86.4%-90.8%。本研究的第二部分設(shè)在5種溫度下,采用十種試劑培養(yǎng)方法來體外培養(yǎng)犬弓首蛔蟲蟲卵,以及對犬弓首蛔蟲蟲卵在自然生活環(huán)境中進(jìn)行體外模擬培養(yǎng)試驗(yàn),結(jié)果顯示,在15℃、25℃、35℃條件下,各體外試劑培養(yǎng)方法組和各模擬試驗(yàn)組的犬弓首蛔蟲蟲卵均有發(fā)育,在5℃、45℃條件下各組均未發(fā)育。十種體外試劑培養(yǎng)方法中,以25℃1molH2SO4培養(yǎng)犬弓首蛔蟲蟲卵孵化最快。模擬實(shí)驗(yàn)中,以25℃清水中的犬弓首蛔蟲蟲卵孵化最快。本研究的第三部分為犬弓首蛔蟲感染性蟲卵的鑒定和犬弓首蛔蟲幼蟲在小雞體內(nèi)移行試驗(yàn)。結(jié)果顯示含兩圈幼蟲期(6字形會動)蟲卵和含兩圈幼蟲期(8字形會動)蟲卵為感染性蟲卵,其幼胚長度均為238-270μm。在小雞感染幼蟲12小時肝臟最早分離出幼蟲,肺臟分離出幼蟲最早時間為感染后24h。本研究為國內(nèi)首次對犬弓首蛔蟲蟲卵在自然環(huán)境進(jìn)行模擬體外培養(yǎng)試驗(yàn),以及對犬弓首蛔蟲感染性蟲卵進(jìn)行的鑒別,在國內(nèi)首次犬弓首蛔蟲幼蟲在小雞體內(nèi)移行試驗(yàn)。本研究為犬弓首蛔蟲的分子生物學(xué)、生物學(xué)特性、臨床試驗(yàn)以及公共衛(wèi)生學(xué)等諸多研究提供了依據(jù)。
[Abstract]:Ascaris canis belongs to the genus Toxocara of the (Ascarididea) family Toxocaridae, a nematode parasitic on the small intestine and stomach of dogs and other animals (including human beings). In order to understand the development of Ascaris canis eggs in vitro, identify their infective eggs, and master the migration of larvae in chickens, a series of related studies were carried out in this study. It provides a great deal of scientific basis for the diagnosis and prevention of Ascaris canis disease. In the first part of this study, 7 cox1 genes of Ascaris canis were sequenced and sequenced. The results showed that the length of cox1 gene of 7 strains of Ascaris canis was 440 BP. The intraspecific similarity of mitochondrial cox1 gene was 96.7 and the interspecific similarity was 86.4-90.8. In the second part of this study, ten reagent culture methods were used to culture the eggs of Ascaris canis in vitro at five temperatures, and the results showed that the eggs of Ascaris canis were cultured in natural living environment in vitro.
