【摘要】：從商品肉鴨場采集鴨糞及墊料,以實(shí)驗(yàn)室保存的沙門菌為宿主菌,采用雙層平板法分離到1株烈性噬菌體。對分離到的噬菌體進(jìn)行純化、效價(jià)測定、裂解譜測定、形態(tài)觀察,并且進(jìn)行了最佳感染復(fù)數(shù)、溫度跟p H值穩(wěn)定性和一步生長曲線的生物學(xué)特性研究。結(jié)果表明,該噬菌體形成的蝕斑均勻透亮,效價(jià)能達(dá)到109PFU/m L以上,并能裂解多株沙門菌菌株,為寬裂解譜噬菌體,透射電鏡顯示,該噬菌體為長尾噬菌體;最佳感染復(fù)數(shù)為0.001;一步生長曲線測定表明,該噬菌體的潛伏期為15 min,暴發(fā)期為30 min,暴發(fā)量約為320;該噬菌體的溫度、p H值耐受性良好;紫外線照射60 min后效價(jià)降低3個(gè)梯度,由此可見該噬菌體分離株可作為潛在開發(fā)的生物消毒劑菌株。
[Abstract]:1 strains of phage were isolated with Salmonella from the commercial meat duck farm, and 1 strains of phage were isolated by the laboratory preserved Salmonella. The isolated phage was purified, titer, cleavage spectrum and morphological observation, and the optimum number of infection, temperature and P H value stability and one step growth curve were produced. The results showed that the plaque formed by the phage was uniform and bright, the potency could reach more than 109PFU/m L, and the multiple strains of Salmonella could be cracked. The phage was a wide lysate phage. The transmission electron microscope showed that the phage was a long tail phage, the optimum number of infection was 0.001, and the one step growth curve showed that the latent period of the phage was 1. 5 min, the outbreak period was 30 min, the outbreak amount was about 320; the temperature of the phage, P H was well tolerated, and the titer decreased by 3 gradient after ultraviolet radiation 60 min, thus the bacteriophage isolate could be used as a potential biodisinfectant strain.
