禽白血病重組毒FJ15TH0株不同感染方式對(duì)SPF雞感染狀態(tài)及組織病毒載量的影響

【摘要】:為揭示禽白血病病毒(ALV)自然重組毒FJ15TH0感染SPF雞群的不同方式對(duì)其感染狀態(tài)和病毒組織分布的影響,本研究設(shè)計(jì)動(dòng)物試驗(yàn)分別模擬垂直傳播和生長(zhǎng)早中期水平傳播,動(dòng)態(tài)檢測(cè)各組病毒血癥、泄殖腔排毒、抗體反應(yīng)及不同內(nèi)臟組織病毒載量,為制定地方雞群中ALV感染的防控和凈化措施提供可靠的流行病學(xué)依據(jù)。結(jié)果表明胚內(nèi)接種模擬垂直傳播組除2只實(shí)驗(yàn)雞外均產(chǎn)生免疫耐受,持續(xù)帶毒排毒而無抗體產(chǎn)生;1日齡和D組均未表現(xiàn)泄殖腔排毒,在接觸病毒后2~3w后C組63.6%實(shí)驗(yàn)動(dòng)物表現(xiàn)一過性病毒血癥,D組未表現(xiàn)病毒血癥,暴露后5~11w期間C組9.1%~25%雞出現(xiàn)一過性血清抗體,D組在暴露后3~5w期間14.3%~42.8%實(shí)驗(yàn)雞產(chǎn)生一過性血清抗體。11w時(shí)剖殺實(shí)驗(yàn)雞,熒光定量RT-PCR檢測(cè)各組各實(shí)驗(yàn)雞不同組織的gp85mRNA及病毒粒子的基因組RNA含量,結(jié)果顯示垂直感染組表現(xiàn)泄殖腔排毒者gp85檢測(cè)均為陽性,該分離株對(duì)腎臟和法氏囊的組織親嗜性最強(qiáng),而對(duì)肝臟和脾臟的親嗜性較低;而同居感染組實(shí)驗(yàn)雞均表現(xiàn)為gp85mRNA檢測(cè)陰性,表明該重組毒水平感染能力較弱。
[Abstract]:In order to reveal the effects of different ways of FJ15TH0 infection on the infection status and tissue distribution of avian leukemia virus (ALV) FJ15TH0, this study designed animal experiments to simulate vertical transmission and horizontal transmission in early and middle stage of growth, respectively. Dynamic detection of viremia, cloaca detoxification, antibody response and viral load in different visceral tissues provided a reliable epidemiological basis for the prevention, control and purification of ALV infection in local chickens. The results showed that immune tolerance was produced in the simulated vertical transmission group except for two experimental chickens, and no cloacal detoxification was observed in group D and no antibody was produced at 1 day after inoculation. 63.6% of the animals in group C showed transient viremia and group D did not show viremia at 2 weeks after exposure to the virus, and 63.6% of the animals in group C showed transient viremia after exposure to the virus. Transient serum antibody was found in group C (9.1g / 25%) during the period of 5w / 11w after exposure. In group D, 14.32.8% chickens produced transient serum antibodies (TSA) during 5 weeks after exposure. Fluorescence quantitative RT-PCR was used to detect the content of gp85 mRNA and genomic RNA of virus particles in different tissues of different chickens in each group. The results showed that the gp85 detection was positive in the patients with vertical infection, and the isolate had the strongest affinity to the kidney and bursa of Fabricius. In the cohabitation group, the gp85 mRNA was negative, which indicated that the recombinant virus level was weak.
