地塞米松對(duì)仔豬腸道黏膜形態(tài)的影響

【摘要】:試驗(yàn)旨在研究地塞米松對(duì)仔豬腸道黏膜形態(tài)的影響,并初步探討地塞米松對(duì)腸道免疫屏障功能的影響以及利用地塞米松建立仔豬腸道損傷模型的可行性,以期為今后研究其他糖皮質(zhì)類激素引起的腸道損傷發(fā)生機(jī)制和防治措施提供理論和科學(xué)依據(jù)。試驗(yàn)選用斷奶1周的健康仔豬30頭,隨機(jī)分為6組,每組5個(gè)重復(fù),試驗(yàn)Ⅰ、Ⅱ、Ⅲ組分別注射地塞米松1、2、3ml,對(duì)照Ⅰ、Ⅱ、Ⅲ組分別注射生理鹽水1、2、3mL;采用HE染色、PAS染色和ELISA檢測(cè)法分別測(cè)定腸道絨毛高度、隱窩深度、絨毛高度與隱窩深度比值、杯狀細(xì)胞、上皮內(nèi)淋巴細(xì)胞和腸黏膜分泌型免疫球蛋白A(sIgA)6個(gè)指標(biāo)。試驗(yàn)結(jié)果表明,地塞米松能抑制仔豬腸絨毛生長(zhǎng),使隱窩深度變深,腸絨毛高度與隱窩深度比值降低,杯狀細(xì)胞、上皮內(nèi)淋巴細(xì)胞數(shù)目和sIgA含量均減少,且劑量越大以上變化越明顯。綜合上述結(jié)果,地塞米松會(huì)影響腸絨毛的發(fā)育和腸道營(yíng)養(yǎng)物質(zhì)的吸收,破壞黏膜免疫屏障功能,且有劑量依賴性。
