6-芐氨基嘌呤對大鼠小腸缺血再灌注損傷的影響

【摘要】:本試驗旨在研究6-芐氨基嘌呤(6-BA)對大鼠小腸缺血再灌注(I/R)損傷的保護(hù)作用。選取80只雄性SD大鼠,隨機(jī)分為4組,即對照組、I/R模型組和低、高劑量6-BA組。低、高劑量6-BA組于術(shù)前3周分別連續(xù)灌胃10、20mg/kg的6-BA,對照組和I/R模型組灌胃同體積的生理鹽水,每天1次。對照組在暴露腸系膜上動脈后不做阻斷;I/R模型組和低、高劑量6-BA組阻斷腸系膜血管30min后再灌注60min。隨后取大鼠空腸組織分別進(jìn)行總超氧化物歧化酶(T-SOD)、谷胱甘肽過氧化物酶(GSH-Px)活性和丙二醛(MDA)含量檢測;采用單細(xì)胞凝膠電泳法檢測細(xì)胞DNA損傷程度,采用免疫組化法檢測半胱氨酸天冬氨酸特異性蛋白酶3(Caspase-3)表達(dá)情況。結(jié)果顯示,與I/R模型組相比,補(bǔ)充10、20 mg/kg的6-BA后,T-SOD、GSH-Px活性顯著升高(P0.05),MDA含量顯著降低(P0.05);小腸細(xì)胞DNA的拖尾現(xiàn)象好轉(zhuǎn),尾部DNA含量和拖尾率都顯著低于I/R模型組(P0.05);Caspase-3陽性表達(dá)細(xì)胞數(shù)量顯著減少(P0.05)。由此可見,10、20 mg/kg的6-BA能有效防護(hù)I/R對小腸的損傷,且20mg/kg的6-BA作用效果尤為明顯。
[Abstract]:The aim of this study was to investigate the protective effect of 6-benzyl aminopurine 6-BA) on intestinal ischemia reperfusion injury in rats. Eighty male Sprague-Dawley rats were randomly divided into 4 groups: control group, I / R model group and low and high dose 6-BA group. The low and high dose 6-BA groups were intragastrically perfused with 10 ~ 20 mg / kg 6-BA for 3 weeks before operation. The control group and the I / R model group were given the same volume of normal saline once a day. The control group did not block the superior mesenteric artery after exposure to the superior mesenteric artery and the model group was low and low. The high dose 6-BA group blocked the mesenteric vessel 30min for 60 minutes after reperfusion. The total superoxide dismutase (T-SODX), glutathione peroxidase (GSH-Px) activity and malondialdehyde (MDA) content were detected in jejunum of rats, and DNA damage was detected by single cell gel electrophoresis. The expression of cysteine aspartate specific proteinase 3 (Caspase-3) was detected by immunohistochemical method. The results showed that compared with the I / R model group, the activity of GSH-Px of T-SODX was significantly increased after 6-BA supplementation for 10 ~ 20 mg/kg, and the MDA content of P0.05 was significantly decreased, and the tail trailing phenomenon of intestinal cells was improved. The tail DNA content and tail dragging rate were significantly lower than those in the I / R model group, and the number of Caspase-3 positive cells was significantly decreased in the I / R model group. It can be seen that 6-BA of 10 ~ 20 mg/kg can effectively protect the small intestine from injury caused by I / R, and the effect of 6-BA of 20mg/kg is especially obvious.
