玉米赤霉烯酮降解菌的篩選及其活性檢測

【摘要】:玉米赤霉烯酮(Zearalenone,ZEN)是危害動物健康的重要真菌毒素之一,筆者旨在篩選出能夠有效降解ZEN的微生物,并對其降解特性以及降解產(chǎn)物的致毒活性進(jìn)行檢測,以期篩選出高效降解ZEN的菌株。以ZEN為唯一碳源進(jìn)行初篩,以菌株發(fā)酵液對ZEN的降解率為復(fù)篩指標(biāo),對降解率最高的菌株進(jìn)行16SrRNA基因測序分析;并對篩選出的菌株對ZEN的降解特性進(jìn)行研究,對其降解產(chǎn)物在MCF-7細(xì)胞上進(jìn)行活性檢測。初篩獲得40株能生長良好的細(xì)菌,復(fù)篩篩選出降解率最高H6菌株,經(jīng)16SrRNA基因測序分析,該菌株為解淀粉芽孢桿菌(Bacillus amyloliquefaciens)。在37℃、180r·min-1、72h時(shí),該菌株發(fā)酵液對1μg·mL~(-1) ZEN的降解率為95.6%,其上清液和菌懸液的降解率分別為68.93%和28.70%,對20μg·mL~(-1) ZEN的降解率為85.8%;菌株上清液經(jīng)蛋白酶K、蛋白酶K+SDS和熱處理后,其降解活性明顯降低;該菌株ZEN的降解產(chǎn)物在MCF-7細(xì)胞上的增殖率顯著降低。結(jié)果表明,本研究篩選出能有效降解ZEN的菌株H6,經(jīng)鑒定判斷為解淀粉芽孢桿菌(B.amyloliquefaciens),該菌株分泌的胞外酶能夠降解ZEN,使其類雌激素毒性顯著降低。
[Abstract]:Zen is one of the most important fungal toxins harmful to animal health. The purpose of this study was to screen out the microorganisms that could effectively degrade Zen, and to detect the degradation characteristics and toxic activity of the degradation products. The aim of this study was to screen the strain that could degrade Zen efficiently. Zen was used as the sole carbon source, the degradation rate of Zen was used as the index of double screening, the 16s rRNA gene of the strain with the highest degradation rate was sequenced, and the degradation characteristics of Zen were studied. The biodegradation products were detected in MCF-7 cells. 40 strains of bacteria which could grow well were screened out, and the highest degradation rate of H6 strain was screened.
