豬濃縮飼料蛋白質(zhì)氧化及不同抗氧化劑抗氧化作用的研究

【摘要】:豬濃縮飼料應(yīng)用較為普遍,其主要組成部分是蛋白質(zhì)飼料,含蛋白質(zhì)較高,通常貯存時(shí)間較長(zhǎng),在儲(chǔ)存過程中蛋白質(zhì)易受各種因素影響而發(fā)生氧化,從而影響其應(yīng)用效果。本研究采集飼料企業(yè)實(shí)際生產(chǎn)的豬濃縮飼料進(jìn)行蛋白質(zhì)氧化狀況分析,并研究蛋白質(zhì)發(fā)生氧化的濃縮飼料對(duì)仔豬的影響;同時(shí)研究不同類型抗氧化劑對(duì)乳豬濃縮飼料蛋白質(zhì)氧化的影響,為保證濃縮飼料的品質(zhì)提供參考依據(jù)。論文包括以下3個(gè)試驗(yàn):試驗(yàn)一調(diào)研分析部分飼料企業(yè)豬濃縮飼料蛋白質(zhì)氧化狀況。選擇江蘇省有代表性的飼料企業(yè)進(jìn)行濃縮飼料樣品采集,根據(jù)生產(chǎn)時(shí)間、儲(chǔ)藏時(shí)間等條件,分為不同的批次,將采集的飼料樣用四分法縮減至200g左右并粉碎過60目,進(jìn)行蛋白質(zhì)氧化指標(biāo)(蛋白質(zhì)羰基、琉基、游離氨基含量)測(cè)定。結(jié)果表明,隨著儲(chǔ)存時(shí)間的延長(zhǎng),濃縮飼料中的蛋白質(zhì)羰基含量逐漸升高(P0.05),蛋白質(zhì)游離巰基、總巰基及游離氨基含量逐漸降低(P0.05),提示蛋白質(zhì)的氧化程度隨著儲(chǔ)存時(shí)間的延長(zhǎng)而升高。試驗(yàn)二研究常規(guī)條件儲(chǔ)存濃縮飼料對(duì)仔豬生長(zhǎng)性能和血清生理生化指標(biāo)的影響。選用健康、體重相近的35日齡斷奶仔豬64頭,隨機(jī)分為2組,每組4重復(fù),每個(gè)重復(fù)8頭豬,日糧處理如下:(1)日糧所用濃縮飼料為4℃低溫儲(chǔ)存45 d;(2)日糧所用濃縮飼料為常規(guī)條件儲(chǔ)存45 d。結(jié)果表明,與低溫儲(chǔ)存組相比,常規(guī)條件儲(chǔ)存組仔豬平均日增重(ADG)和平均日采食量(ADIF)顯著降低(P0.05),仔豬血清谷丙轉(zhuǎn)氨酶(ALT)活性及白蛋白(ALB)含量顯著降低(P0.05),血清尿素氮(UN)含量顯著升高(P0.05)。試驗(yàn)三研究不同類型抗氧化劑對(duì)濃縮飼料中蛋白質(zhì)氧化的影響。選用乳豬濃縮飼料為試驗(yàn)材料,處理如下:(1)AO1組不添加任何抗氧化劑;(2)AO2組添加固體乙氧基喹淋;(3)AO3組添加液體乙氧基喹淋;(4)AO4組添加固體和液體乙氧基喹啉。添加劑量均為150g/t乙氧基喹淋。將各處理飼料樣品分別置于室溫25 ℃和模擬夏季高溫35 ℃培養(yǎng)箱內(nèi)儲(chǔ)存不同時(shí)間(0d,10 d,20 d,30 d和60 d)。結(jié)果表明,隨著儲(chǔ)存時(shí)間的延長(zhǎng)各組濃縮飼料蛋白質(zhì)羰基含量顯著升高(P0.05),巰基含量顯著降低(P0.05),游離氨基含量沒有顯著變化(P0.05),NSI及蛋白質(zhì)體外消化率顯著降低(P0.05);乙氧基喹淋具備一定的抗氧化作用,儲(chǔ)存時(shí)間在20 d以前4組之間蛋白質(zhì)氧化程度沒有顯著差異,儲(chǔ)存30 d以后,液體乙氧基喹淋的抗氧化效果顯著優(yōu)于固體乙氧基喹啉(P0.05),固、液體乙氧基喹啉各半使用的抗氧化效果不穩(wěn)定。
[Abstract]:Porcine concentrated feed is widely used and its main component is protein feed, which contains high protein and usually has a long storage time. During storage, protein is easily oxidized by various factors, thus affecting its application effect. In this study, the protein oxidation status of pig concentrated feed produced by feed enterprises was analyzed, and the effect of protein oxidized concentrated feed on piglets was studied. At the same time, the effects of different antioxidants on protein oxidation in concentrated feed of suckling pigs were studied in order to guarantee the quality of concentrated feed. The thesis includes the following three experiments: experiment 1: investigation and analysis of protein oxidation status of pig concentrate feed in some feed enterprises. The representative feed enterprises in Jiangsu Province were selected to collect concentrated feed samples. According to the production time, storage time and other conditions, the samples were divided into different batches, and the collected feed samples were reduced to about 200g by the four-part method and crushed over 60 mesh. Protein oxidation index (protein carbonyl, Ryukyl, free amino content) was determined. The results showed that the content of protein carbonyl in concentrated feed increased gradually with the prolongation of storage time, and the protein free sulfhydryl group increased gradually. The content of total sulfhydryl group and free amino group decreased gradually, suggesting that the oxidation degree of protein increased with the prolongation of storage time. The second experiment was to study the effects of conventional storage of concentrated feed on growth performance and serum physiological and biochemical indexes of piglets. Sixty-four weaned piglets of 35 days of similar body weight were randomly divided into 2 groups with 4 replicates in each group and 8 piglets in each replica.
