雞血藤總黃酮對豬圓環(huán)病毒2型感染小鼠脾臟氧化應(yīng)激的影響

【摘要】:試驗旨在探討雞血藤總黃酮(TFSD)對豬圓環(huán)病毒2型(PCV2)感染小鼠脾臟氧化應(yīng)激的影響。將70只昆明小鼠隨機分為7組,對照組、TFSD組(100mg/kg體重)、PCV2組、PCV2+維生素C(VC)組、PCV2+不同濃度TFSD(25、50和100mg/kg體重)組,每組10只,連續(xù)3d采用灌胃和腹腔注射PCV2病毒液的方法建立小鼠氧化應(yīng)激模型,第4~6天每天按上述分別灌胃給予生理鹽水、VC或TFSD。第7天剖殺小鼠,取脾臟分析黃嘌呤氧化酶(XOD)、髓過氧化物酶(MPO)和超氧化物歧化酶(SOD)活力,并檢測還原型谷胱甘肽(GSH)和氧化型谷胱甘肽(GSSG)水平,計算GSH/GSSH。結(jié)果顯示,PCV2感染小鼠后,脾臟中XOD與MPO的活力及GSSG水平顯著上升(P0.05),SOD活力、GSH水平及GSH/GSSG顯著下降(P0.05)。TFSD處理小鼠脾臟的SOD活力、GSH水平和GSH/GSSG比值均顯著高于PCV2組(P0.05),而XOD、MPO活力和GSSG水平則顯著低于PCV2組(P0.05),對PCV2引起的氧化應(yīng)激相關(guān)酶活力與相關(guān)分子水平變化的抑制作用優(yōu)于VC。結(jié)果表明,雞血藤總黃酮對PCV2誘導(dǎo)的小鼠脾臟氧化應(yīng)激有良好的調(diào)節(jié)作用。
[Abstract]:The purpose of this study was to investigate the effect of TFSD on oxidative stress in spleen of mice infected with porcine circovirus type 2 (PCV2). 70 Kunming mice were randomly divided into 7 groups. The control group was divided into two groups: the control group (100 mg / kg BW) and the PCV2 vitamin C (VC2) group (10 rats in each group) with different concentrations of TFV2, TFSDD, 2550 and 100mg/kg body weight (n = 10 in each group), and 10 mice in each group were divided into two groups, the control group (n = 10) and the control group (n = 10). The oxidative stress model of mice was established by intraperitoneal injection of PCV2 virus solution and intragastric perfusion for 3 consecutive days. The mice were given normal saline VC or TFSD respectively daily on the 4th day of 6 days. On the 7th day, the mice were dissected and the activities of XODO, MPO and SOD were analyzed, and the GSH) and GSSGG levels of GSH and GSSG were measured. The GSH / GSSH was calculated. The results showed that PCV2 infected mice, The activities of XOD, MPO and GSSG in spleen increased significantly. The activity of SOD and GSH of GSH / GSSG decreased significantly in the spleen of mice treated with P0.05. TFSD. The activity of SOD and the ratio of GSH / GSSG in spleen were significantly higher than those in group PCV2, while the activity of XODMPO and the level of GSSG in PCV2 group were significantly lower than those in group PCV2. The inhibitory effect of P0.05 on the activity of oxidative stress-related enzymes and the changes of related molecules induced by PCV2 was superior to that of VC. The results showed that total flavone could regulate the oxidative stress induced by PCV2 in mice.
