3號中草藥添加劑對仔豬免疫功能影響的研究

【摘要】:中草藥具有毒副作用小,綠色無殘留的特點,已成為國內飼料添加劑研究領域的熱點,尤其是中草藥添加劑提高動物機體抗病能力的研究,越來越受到人們的關注。為探討3號中草藥添加劑對仔豬部分主要免疫指標的影響,為提高仔豬抗病力和養(yǎng)豬業(yè)的綠色健康發(fā)展提供科學依據(jù)。試驗隨機選取15日齡左右經(jīng)檢測Mhp抗體為陰性的仔豬60頭,隨機分為對照組、中草藥組、疫苗組、中草藥+疫苗組,連續(xù)飼喂3號中草藥10d,分別進行了3號中草藥添加劑對仔豬支原體肺炎滅活疫苗免疫效果,部分血常規(guī)和血液生化指標,血清中IL-2、IL-4、IFN-γ的含量,仔豬T淋巴細胞亞群和增殖影響的試驗。結果如下:1、3號中草藥添加劑對仔豬支原體肺炎滅活疫苗免疫效果的影響為探討3號中草藥添加劑對仔豬滅活疫苗免疫效果的影響,試驗對疫苗組和中草藥+疫苗組的仔豬15日齡首免安百克滅活疫苗,并與30日齡二免,分別在實驗前、30日齡和50日齡采集血清,采用ELISA方法檢測豬支原體肺炎抗體水平。結果:30日齡和50日齡時,疫苗組和中草藥+疫苗組的S/P值均高于空白對照組和中草藥(P0.01);中草藥+疫苗組的S/P值顯著高于疫苗組(P0.05)。顯示:3號中草藥添加劑能夠提高仔豬支原體肺炎滅活疫苗免疫效果。2、3號中草藥添加劑對仔豬部分血常規(guī)和血液生化指標的影響為探討3號中草藥添加劑對仔豬部分血常規(guī)和血清生化指標的影響,試驗分別于實驗前、30日齡和50日齡采血,采用全自動動物血細胞分析儀和血液生化分析儀進行檢測,結果:30日齡時,與對照組相比:中草藥組、疫苗組、中草藥+疫苗組的WBC分別提高了4.75%、8.72%、18.62%(P0.05);LYM分別提高了9.83%、15.08%、25.57%(P0.05);Gran分別提高了15.12%、21.00%、36.12%;RBC分別提高了8.36%、2.85%、9.12%;TP分別提高了15.81%(P0.05)、6.67%、16.85%(P0.05);ALB分別提高了6.37%、-0.74%、10.61%;GLO分別提高了38.71%(P0.05)、24.61%、32.00%(P0.05)。50日齡時,與對照組相比:中草藥組、疫苗組、中草藥+疫苗組的WBC分別提高了8.39%、34.54%、36.45%;LYM分別提高了28.80%、12.58%、31.78%;Gran分別提高了12.23%、0.53%、29.25%;RBC分別提高了11.45%、7.21%、27.23%(P0.05);TP分別提高了14.87%(P0.05)、12.30%(P0.05)、23.45%(P0.05);ALB分別提高了11.08%、6.59%、14.07%(P0.05);GLO分別提高了22.81%、24.27%、43.10%(P0.05)。表明:3號中草藥能夠提高仔豬外周血中白細胞、淋巴細胞、紅細胞的數(shù)量和清中總蛋白、白蛋白和球蛋白的含量。3、3號中草藥添加劑對仔豬血清中IL-2、IL-4、IFN-γ含量的影響為探討3號中草藥添加劑對仔豬血清中IL-2、IL-4、IFN-γ含量的影響,試驗采用ELISA試劑盒進行了檢測,結果:30日齡時,與空白對照組相比:中草藥組、疫苗組、中草藥+疫苗組IL-2的濃度分別提高了0.92%、4.14%、5.32%(P0.05);IL-4的濃度分別提高了3.44%、14.22%(P0.05)、12.18%(P0.05);IFN-γ的濃度分別提高了4.11%、6.55%、21.12%(P0.05)。50日齡時,與空白對照組相比:中草藥組、疫苗組、中草藥+疫苗組IL-2的濃度分別提高了4.10%(P0.05)、1.05%、12.75%(P0.05);IL-4的濃度分別提高了7.63%、7.42%、8.54%;IFN-γ的濃度分別提高了4.15%、4.06%、12.44%(P0.05)。說明:3號中草藥添加劑提高了仔豬血清中IL-2、IL-4、IFN-γ的濃度,尤其是顯著提高了中草藥+疫苗組的IL-2、IL-4、IFN-γ含量。4、3號中草藥添加劑對仔豬T淋巴細胞亞群及T淋巴細胞體外增殖的影響為探討3號中草藥添加劑對仔豬T淋巴細胞亞群及T淋巴細胞體外增殖的影響,試驗采用流式細胞術檢測仔豬外周血T淋巴細胞亞群CD4~+、CD8~+的百分比變化;采用CCK-8法進行了T淋巴細胞體外增殖試驗。結果:30日齡時,與對照組相比:中草藥組、疫苗組、中草藥+疫苗組的CD4~+T細胞百分比分別提高了8.96%、16.91%、26.24%;CD8~+T細胞百分比分別提高了3.33%、4.62%、7.02%(P0.05);CD4~+/CD8~+的值分別增長6.88%、11.85%、19.41%(P0.05);T淋巴細胞刺激指數(shù)(SI)分別增長了5.47%,3.48%和10.02%(P0.05)。50日齡時,與對照組相比:中草藥組、疫苗組、中草藥+疫苗組的CD4~+T細胞的百分比分別提高了13.43%、10.47%、33.30%(P0.05);CD8~+T細胞百分比分別提高了2.42%、5.00%、-2.21%;CD4~+/CD8~+的值分別提高了14.87%、6.49%、32.68%(P0.05);T淋巴細胞刺激指數(shù)(SI)分別增長了6.90%,2.24%和9.80%(P0.05)。表明:3號中草藥添加劑能夠提高仔豬外周血中CD4~+T細胞百分比、CD4~+/CD8~+的比例和T淋巴細胞體外增殖能力,尤其是能顯著提高中草藥+疫苗組的CD4~+T細胞、CD8~+T細胞的百分比以及CD4~+/CD8~+的比例和T淋巴細胞刺激指數(shù)(SI)。因此,3號中草藥能夠提高仔豬支原體滅活疫苗的免疫效果,血液中白細胞、紅細胞和淋巴細胞的數(shù)量,血清中總蛋白、白蛋白、球蛋白、IL-2、IL-4、IFN-γ的含量,CD4~+T細胞的百分比和CD4~+/CD8~+的值和T淋巴細胞的體外增殖能力,從而提高了仔豬的免疫功能和抗病能力。
[Abstract]:Chinese herbal medicine has the characteristics of small toxic and side effects and no residue of green, which has become a hot spot in the research field of domestic feed additives, especially the study on improving the disease resistance of animal body by Chinese herbal medicine additive, which has attracted more and more attention. A scientific basis was provided for the green and healthy development of the disease power and the pig industry. The test randomly selected 60 piglets with negative Mhp antibody for 15 days of age and randomly divided into the control group, the Chinese herbal medicine group, the vaccine group, the Chinese herbal medicine + vaccine group, and the continuous feeding of the Chinese herbal medicine 10d, which was used to inactivate the piglet mycoplasma pneumonia inactivated vaccine. The immune effect, some blood routine and blood biochemical indexes, the content of IL-2, IL-4, IFN- gamma in the serum, the effects of the T lymphocyte subsets and proliferation of piglets. The results are as follows: the effect of the Chinese herbal additive on the immune effect of the piglet mycoplasma pneumonia inactivated vaccine is the shadow of the effect of the 3 Chinese herbal additive on the inactivated vaccine of piglet inactivated vaccine. The percentage increased by 13.43%, 10.47%, 33.30% (P0.05), and the percentage of CD8~+T cells increased by 2.42%, 5%, and -2.21%, respectively, and the value of CD4~+/CD8~+ increased by 14.87%, 6.49%, 32.68% (P0.05), and the T lymphocyte stimulation index (SI) increased by 6.90%, 2.24% and 9.80% (P0.05), respectively. It indicated that the herb additive in 3 was able to increase the peripheral blood of piglets. The percentage of CD4~+T cells, the proportion of CD4~+/CD8~+ and the proliferation ability of T lymphocytes in vitro, especially the CD4~+T cells, the percentage of CD8~+T cells and the proportion of CD4~+/CD8~+ and the T lymphocyte stimulation index (SI) in the Chinese herbal + vaccine group. Therefore, 3 Chinese herbal medicine can improve the immune effect of the piglet inactivated vaccine. The number of leukocytes, red cells and lymphocytes in the liquid, the content of total protein, albumin, globulin, IL-2, IL-4, IFN- gamma in serum, the percentage of CD4~+T cells and the value of CD4~+/CD8~+ and the ability of T lymphocyte proliferation in vitro, thus improve the immune function and disease resistance of piglets.
