蛋氨酸缺乏對(duì)蛋雞產(chǎn)蛋后期生產(chǎn)性能、血清游離氨基酸含量和肝臟蛋氨酸代謝相關(guān)基因表達(dá)的影響

【摘要】:本試驗(yàn)旨在研究蛋氨酸缺乏對(duì)蛋雞產(chǎn)蛋后期生產(chǎn)性能、血清游離氨基酸含量和肝臟蛋氨酸代謝相關(guān)酶基因表達(dá)的影響。選取180只62周齡的海蘭灰蛋雞,根據(jù)產(chǎn)蛋率均勻分成3組(每組6個(gè)重復(fù),每個(gè)重復(fù)10只):對(duì)照組飼喂蛋氨酸水平為0.33%的飼糧,蛋氨酸缺乏組分別飼喂蛋氨酸水平為0.21%和0.27%的飼糧,試驗(yàn)期90d。結(jié)果表明:1)飼糧蛋氨酸缺乏顯著降低了蛋雞的平均日采食量、產(chǎn)蛋率、平均日產(chǎn)蛋重、平均蛋重和不合格蛋率(P0.05),顯著提高了料蛋比(P0.05)。2)蛋氨酸缺乏顯著降低蛋雞血清中蛋氨酸的含量(P0.05)。0.21%蛋氨酸組蛋雞血清中絲氨酸、甘氨酸和丙氨酸的含量顯著高于其他2組(P0.05)。0.27%蛋氨酸組蛋雞血清中纈氨酸、異亮氨酸和精氨酸的含量顯著低于對(duì)照組(P0.05),脯氨酸的含量顯著低于0.21%蛋氨酸組(P0.05)。3)與對(duì)照組相比,0.21%蛋氨酸組蛋雞肝臟中DNA甲基轉(zhuǎn)移酶1、N6-甲基腺苷(m6 A)甲基轉(zhuǎn)移酶3(METTL3)和m6 A甲基轉(zhuǎn)移酶14的表達(dá)量顯著提高(P0.05),0.27%蛋氨酸組蛋雞肝臟中METTL3的表達(dá)量顯著提高(P0.05)。4)與對(duì)照組相比,0.21%蛋氨酸組蛋雞肝臟中甲硫氨酸腺苷轉(zhuǎn)移酶1a、5-甲基四氫葉酸-同型半胱氨酸甲基轉(zhuǎn)移酶(MTR)和胱硫醚-β-合成酶的表達(dá)量顯著提高(P0.05),0.27%蛋氨酸組蛋雞肝臟中MTR的表達(dá)量顯著提高(P0.05)。蛋氨酸缺乏對(duì)蛋雞肝臟甲硫氨酸腺苷高半胱氨酸酶和甜菜堿高半胱氨酸甲基轉(zhuǎn)移酶的表達(dá)量無(wú)顯著影響(P0.05)。由以上結(jié)果可知:蛋雞飼糧高水平的蛋氨酸缺乏會(huì)降低蛋雞的生產(chǎn)性能,可能與蛋氨酸缺乏改變蛋氨酸代謝途徑,影響DNA和RNA的甲基化過(guò)程有關(guān)。
[Abstract]:The purpose of this study was to study the effects of methionine deficiency on the performance of laying hens, the content of serum free amino acids and the expression of enzymes related to methionine metabolism in liver. 180 Hailan gray laying hens aged 62 weeks were divided into 3 groups according to laying rate (6 replicates in each group, 10 in each replica). The control group was fed a diet of 0.33% methionine level. Methionine deficiency group was fed with 0.21% and 0.27% methionine levels for 90 days. The results showed that dietary methionine deficiency significantly decreased the average daily feed intake, laying rate and average daily egg weight of laying hens. The average egg weight and unqualified egg rate (P0.05N) significantly increased the ratio of feed to egg (P0.05U. 2) Met deficiency significantly decreased the content of methionine in the serum of laying hens and the serine content in the serum of laying hens was significantly decreased by P0.05- 0.21% methionine. The contents of glycine and alanine were significantly higher than those in the other two groups. The contents of isoleucine and arginine were significantly lower than those of the control group (P0.05A and proline) the content of proline was significantly lower than that of 0.21% methionine group (P0.05. 3) compared with the control group, the content of DNA methyltransferase (1N6-methyladenosine) methyltransferase (3METTL3) in the liver of laying hens with 0.21% methionine was significantly lower than that in the control group. The expression of m6A methyltransferase 14 and m6A methyltransferase 14 significantly increased the expression of METTL3 in the liver of laying hens with 0.27% methionine.) compared with the control group, the expression of methionine adenosyltransferase 1 aa5-methyltetrahydrofolate in the liver of 0.21% methionine group was significantly higher than that of the control group. The expression of homocysteine methyltransferase (MTR-) and cystathithione- 尾-synthase (P0.05- 尾-synthase) increased significantly in the liver of laying hens treated with 0.27% methionine. Methionine deficiency had no significant effect on the expression of methionine adenosine homocysteinase and betaine homocysteine methyltransferase in the liver of laying hens. The results showed that the high level of methionine deficiency in laying diet could decrease the performance of laying hens, which might be related to the effect of methionine deficiency on the metabolic pathway of methionine and the methylation of DNA and RNA.
