【摘要】：內(nèi)參基因表達(dá)穩(wěn)定性的高低將決定著實時熒光定量PCR(Quantitative Real-time PCR,qRT-PCR)分析結(jié)果的可靠程度。本實驗旨在篩選不同濃度氨基葡萄糖(glucosamine,GluN)處理下產(chǎn)蛋后期籠養(yǎng)蛋雞(Gallus gallus domesticus)中穩(wěn)定可靠的內(nèi)參基因,以確保產(chǎn)蛋后期蛋雞基因表達(dá)分析結(jié)果的可靠性。實驗選用500日齡產(chǎn)蛋后期海蘭灰蛋雞作為研究對象,對照組飼喂玉米-豆粕型基礎(chǔ)飼糧,實驗組分別在基礎(chǔ)飼糧中添加0.4%、0.6%和0.8%GluN,運用q RT-PCR技術(shù)對核糖體蛋白S2(ribosomal protein S2,RPS2)、肌動蛋白(β-Actin)、3-磷酸甘油醛脫氫酶(glyceraldehyde-3-phosphate dehydrogenase,GAPDH)、羥甲基膽色烷合成酶(hydroxymethyl biliary synthetase,HMBS)、端粒結(jié)合蛋白(TATA-box binding protein,TBP)、次黃嘌呤鳥嘌呤磷酸核糖轉(zhuǎn)移酶1(hypoxanthine phosphoribosyltransferase 1,HPRT1)、微管蛋白(tubulin beta class,TUBB)、琥珀酸脫氫酶復(fù)合物亞單位A(succinate dehydrogenase complex flavoprotein subunit A,SDHA)、核糖體蛋白L4(ribosomal protein L4,RPL4)和微球蛋白(beta-2 microglobulin,B2M)等10個內(nèi)參基因在不同組織中(心、肝、肺、腎、十二指腸、脛骨、胰和子宮)的表達(dá)情況進(jìn)行分析,同時借助循環(huán)閾值法(cycle threshold,Cq)、Delta循環(huán)閾值法(Delta cycle threshold,Delta CT)和ge Norm、Norm Finder、Ref Finder軟件對每個內(nèi)參基因的表達(dá)穩(wěn)定性進(jìn)行評估。結(jié)果顯示,在利用原始Cq平均值分析時發(fā)現(xiàn),同一內(nèi)參基因在不同處理和不同組織中的表達(dá)量不同,說明所選擇的10個候選內(nèi)參基因在組織間存在一定的表達(dá)差異性;Cq值法分析顯示,RPS2基因的表達(dá)穩(wěn)定性最好;Delta CT法分析發(fā)現(xiàn),不同的GluN水平處理條件下,內(nèi)參基因的表達(dá)穩(wěn)定性并不相同,綜合來看TUBB、β-Actin和RPS2基因的穩(wěn)定性相對較好;ge Norm程序在不同的GluN水平下,依次給出了TBP/RPS2(1.08)、RPS2/β-Actin(0.88)、TBP/TUBB(1.16)和RPS2/HMBS(0.77)表達(dá)穩(wěn)定性內(nèi)參基因組合,由此說明TBP、β-Actin和RPS2基因的穩(wěn)定性較好;Norm Finder程序分析顯示RPS2和HMBS基因表達(dá)穩(wěn)定性較好;Ref Finder在線分析軟件顯示,0.0%GluN和0.8%GluN條件下RPS2基因穩(wěn)定性最高。綜合本研究的分析結(jié)果,RPS2和TBP這樣的內(nèi)參基因適合用于產(chǎn)蛋后期籠養(yǎng)蛋雞基因定量表達(dá)分析,而內(nèi)參基因RPL4的表達(dá)穩(wěn)定性最差,最不適合用于蛋雞的定量分析內(nèi)參,本實驗結(jié)果對GluN處理下矯正目的基因的表達(dá)提供了理論依據(jù)。
[Abstract]:The stability of internal reference gene expression will determine the reliability of real-time fluorescence quantitative PCR(Quantitative Real-time qRT-PCRR analysis. The aim of this study was to screen the stable and reliable internal reference genes of different concentrations of glucosamine gallus (GluN) in the later laying stage of cage laying hens, so as to ensure the reliability of the results of gene expression analysis in the late laying stage of laying hens. In the experiment, 500 days old Hailan grey laying hens were used as the research object, and the control group was fed with corn-soybean meal diet. In the experimental group, 0. 4% and 0. 8 GluNs were added to the basal diet, respectively. The ribosomal protein S2(ribosomal protein S2 / RPS2, 尾 -Actinine 3-glyceraldehyde-3-phosphate dehydrogenase, hydroxymethyl biliary synthetase HMBSN, telomeric binding protein TATA-box binding protein, hypoxanthine were detected by Q RT-PCR technique. Ten internal reference genes of guanine phosphate ribonucleosyltransferase (1(hypoxanthine phosphoribosyltransferase 1), tubulin beta (tubulin beta), succinate dehydrogenase complex subunit (A(succinate dehydrogenase complex flavoprotein subunit), ribosomal protein L4(ribosomal protein L4 (RPL4) and microglobulin beta-2 microglobulinB2M (B2M) were found in different tissues. The expression of liver, lung, kidney, duodenum, tibia, pancreas and uterus were analyzed, and the stability of expression of each internal reference gene was evaluated by cycle thresholdc threshold method, Delta cycle threshold method and ge NormNorm FinderRef Finder software. The results showed that the expression of the same internal reference gene was different in different treatments and tissues by using the original Cq mean value analysis. The analysis of Cq value showed that the stability of RPS2 gene expression was the best. The results of Delta CT analysis showed that under different GluN levels, the expression of RPS2 gene was more stable than that of control group. The stability of Tupb, 尾 -Actin and RPS2 genes was relatively good in different GluN levels, and the relative stability of TBP / 尾 -Actin and RPS2 genes was obtained in turn at different GluN levels, and the expression stability of TBP / 尾 -Actinine 0.88% TBP- TUBB1.16) and RPS2 / HMBS0.77) were obtained respectively, and the relative stability of Tupb, 尾 -Actin-Actin and RPS2 genes were compared with that of RPS2HMBS0.77. The results showed that the stability of RPS2, 尾 -Actin and RPS2 genes was better than that of normal Finder program. The stability of RPS2 and HMBS gene expression was better than that of Ref Finder software. The stability of RPS2 gene was the highest under the condition of GluN and 0.8%GluN. According to the results of this study, RPS2 and TBP are suitable for quantitative analysis of gene expression in cage laying hens, while RPL4 is the least stable, and is the least suitable for quantitative analysis of internal parameters in laying hens. The results of this study provide a theoretical basis for the expression of the target gene under GluN treatment.
