【摘要】：為確診海南省某雞場(chǎng)疑似心包積液綜合征病例,于2016年5月從海南省某雞場(chǎng)送檢的病雞中采集病料(肝臟、心臟),研磨,通過卵黃囊接種7日齡SPF雞胚,并進(jìn)行雞胚病變特征觀察、禽腺病毒PCR特異性檢測(cè)、基因測(cè)序和Blast序列比對(duì)。結(jié)果表明,接種雞胚后5d開始死胚,剖檢可見胚體發(fā)育受阻,羽毛稀少,體表潮紅,雞胚皮膚表面有出血點(diǎn);在病料研磨液、尿囊液及胚體肝臟中均檢測(cè)到大小約為884bp目的片段,將分離病毒命名為HN1605;hexon基因Blast序列比對(duì)及遺傳進(jìn)化分析結(jié)果表明,分離毒株與2015年山東和湖北報(bào)道的Ⅰ亞群腺病毒血清4型(FAV-4)SDSX、SDSX-15、SDNY2-15、HB1510和HBLF-15分離株親緣關(guān)系最近。本研究表明,此次心包積液綜合征病原是Ⅰ亞群禽腺病毒血清4型病毒,該病毒依然危害著我國(guó)雞群,疫情不斷擴(kuò)大,并呈現(xiàn)由我國(guó)北方地區(qū)向南方地區(qū)擴(kuò)散的趨勢(shì)。
[Abstract]:In order to confirm the suspected case of pericardial effusion syndrome in a chicken farm in Hainan province, the infected chicken samples (liver, heart, ground) were collected from a chicken farm in Hainan province in May 2016. The embryo of 7 day old SPF chicken was inoculated with yolk sac. The pathological features of chicken embryo were observed, PCR specificity of avian adenovirus, gene sequencing and Blast sequence alignment were detected. The results showed that the embryo began to die 5 days after inoculation, the embryo development was blocked, the feathers were rare, the body surface was flushed, and there were bleeding spots on the skin of chicken embryo. 884bp fragments were detected in allantoic fluid and embryoid liver. The isolated virus was named HN1605hexon gene Blast sequence alignment and genetic evolution analysis. The isolates were most closely related to the serotype of adenovirus subgroup 鈪，
