治療奶牛子宮內(nèi)膜炎中藥復(fù)方泡騰栓的研究

【摘要】:奶牛子宮內(nèi)膜炎是造成奶牛不孕的主要原因之一,對奶牛養(yǎng)殖業(yè)有著嚴(yán)重的危害,研究出治療該病的良方迫在眉睫。應(yīng)用中藥組方不僅能夠保證療效,還能減小藥物殘留,改善菌株耐藥性的問題。本研究根據(jù)中獸醫(yī)理論選擇多種中草藥,采用正交試驗,對奶牛子宮內(nèi)膜炎的4種主要致病菌進行體外抑菌試驗,根據(jù)中草藥體外抑菌結(jié)果,篩選由雙花、黃芩、等8味中草藥,組合而成得到抑菌作用比較明顯的復(fù)方。泡騰栓具有刺激性小、使用方便,在與體液接觸后可產(chǎn)生大量泡沫,能夠深入子宮黏膜的深處,將藥物釋放到細菌的感染之處,最大限度的發(fā)揮藥效。本試驗用正交法對泡騰栓基質(zhì)進行篩選,以篩選的中藥組方為主藥,制備中藥泡騰栓。對制備的泡騰栓的外觀、重量差異、融變時限、熔點范圍、變形溫度、p H等理化性狀進行了檢查和測定;用中藥泡騰栓對4種致病菌進行了體外抑菌試驗。結(jié)果表明,中藥泡騰栓的最佳配比為:PEG-6000 1.4g,碳酸氫鈉0.26g,檸檬酸0.24g,吐溫-80 0.03g,主藥0.404g;中藥泡騰栓的理化性狀均符合中國藥典(2010版)規(guī)定;中藥泡騰栓對4種致病菌的抑制效果與原藥相近。采用高效液相色譜(HPLC)法對中藥泡騰栓中的幾種單味中藥的有效成分綠原酸和黃芩苷進行了含量測定,考察了產(chǎn)品質(zhì)量的穩(wěn)定性和可控性。綠原酸和黃芩苷分別在3.2-25.6μg/mL和20.8-62.4μg/mL范圍內(nèi)有著良好的線性關(guān)系;二者的精密度和重現(xiàn)性均為良好;綠原酸和黃芩苷在每粒中藥泡騰栓中的含量分別為0.00291g和0.01348g;綠原酸和黃芩苷的平均加樣回收率分別為99.46%和99.23%。結(jié)果表明,該方法對于質(zhì)量控制是穩(wěn)定、可控的。
[Abstract]:Endometritis is one of the main causes of infertility in dairy cows. The application of traditional Chinese medicine can not only guarantee the curative effect, but also reduce the drug residue and improve the drug resistance of strains. In this study, according to the theory of Chinese veterinarian, many kinds of Chinese herbal medicines were selected, and four main pathogenic bacteria of endometritis of dairy cows were tested by orthogonal test in vitro. According to the results of bacteriostasis of Chinese herbal medicine in vitro, Scutellaria baicalensis were selected from Shuanghua and Scutellaria. 8 Chinese herbal medicines, combined to obtain a more obvious bacteriostatic effect of compound. Effervescent suppository has small irritation, easy to use, can produce a large number of foam after contact with body fluid, can go deep into the uterine mucosa, release the drug to the bacterial infection, and give full play to the drug effect. In this experiment, the matrix of effervescent suppository was screened by orthogonal method, and the Chinese medicine effervescent suppository was prepared. The physical and chemical properties of the effervescent suppository, such as appearance, weight difference, melting time, melting point range, deformation temperature and pH, were examined and determined, and four pathogenic bacteria were tested in vitro with Chinese medicine effervescent suppository. The results showed that the optimum proportion of Chinese medicine effervescent suppository was: 1: PEG-6000, sodium bicarbonate 0.26g, citric acid 0.24g, Tween -800.03g, main drug 0.404g.The physicochemical properties of Chinese medicine effervescent suppository were in accordance with the Chinese Pharmacopoeia 2010 edition. The inhibitory effect of traditional Chinese medicine effervescent suppository on 4 kinds of pathogenic bacteria was similar to that of original medicine. High performance liquid chromatography (HPLC) was used to determine the contents of Lv Yuan acid and baicalin in several kinds of Chinese medicine effervescent suppositories. The stability and controllability of the products were investigated. The linear range of Lv Yuan acid and baicalin was 3.2-25.6 渭 g/mL and 20.8-62.4 渭 g/mL, respectively. The contents of Lv Yuan acid and baicalin in each Chinese medicine effervescent suppository were 0.00291g and 0.01348g, respectively, and the average recoveries of Lv Yuan acid and baicalin were 99.46% and 99.23g, respectively. The results show that the method is stable and controllable for quality control.
