黃芩苷對(duì)熱應(yīng)激小鼠睪丸氧化損傷的保護(hù)作用

【摘要】:哺乳動(dòng)物各種生理功能中,生殖功能最易受熱應(yīng)激損傷。熱應(yīng)激使組織產(chǎn)生氧化應(yīng)激,破壞雄性動(dòng)物精子生成,從而影響繁殖效率。為探究黃芩苷對(duì)熱應(yīng)激小鼠(Mus musculus)睪丸損傷的影響,探討黃芩苷的抗氧化作用機(jī)制,本研究將小鼠分成4組,常溫對(duì)照組(C組)、黃芩苷組(C+B組)、熱應(yīng)激組(H組)和熱應(yīng)激加黃芩苷組(H+B組)。C組和H組小鼠腹腔注射生理鹽水,C+B組和H+B組小鼠腹腔注射黃芩苷(50 mg/kg),連續(xù)注射7 d,H組與H+B組于第8天熱應(yīng)激(41℃)2 h,熱應(yīng)激結(jié)束后立即用斷頸法處死所有處理組和對(duì)照組小鼠,取睪丸及附睪檢測(cè)臟器系數(shù)和制備睪丸組織切片,睪丸組織用于檢測(cè)氧化損傷指標(biāo)丙二醛(malondialdehyde,MDA)的含量及超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽過(guò)氧化物酶(glutathione peroxidase,GSH-Px)和過(guò)氧化氫酶(catalase,CAT)的活力。末端脫氧核苷酸轉(zhuǎn)移酶檢測(cè)法(Td T-mediated d UTP nick-end labeling,TUNEL)法檢測(cè)睪丸組織細(xì)胞的凋亡。Western blot檢測(cè)睪丸組織內(nèi)Fas/Fas L蛋白的表達(dá)。結(jié)果顯示,熱應(yīng)激能夠顯著降低睪丸和附睪臟器系數(shù)(P0.05)并使睪丸組織結(jié)構(gòu)發(fā)生病理性損傷,而黃芩苷可以顯著緩解熱應(yīng)激引起的睪丸臟器系數(shù)降低(P0.05)和組織損傷。與熱應(yīng)激組相比,熱應(yīng)激加黃芩苷組小鼠睪丸組織中MDA含量顯著降低,SOD、CAT和GSH-Px活力則顯著增加(P0.05)。黃芩苷組與熱應(yīng)激加黃芩苷組相比則無(wú)顯著性差異。熱應(yīng)組小鼠睪丸組織細(xì)胞凋亡率與對(duì)照組相比顯著增加(P0.05),而熱應(yīng)激黃芩苷組與熱應(yīng)激組相比,小鼠睪丸細(xì)胞凋亡率則顯著降低(P0.05)。此外,與對(duì)照組相比,熱應(yīng)激可以顯著增加小鼠睪丸組織Fas(P0.05)和/Fas L(P0.01)蛋白表達(dá),而黃芩苷能顯著降低熱應(yīng)激引起的小鼠睪丸組織中Fas/Fas L蛋白表達(dá)(P0.05)。本研究結(jié)果表明,黃芩苷能減少受熱應(yīng)激時(shí)小鼠睪丸組織因氧化損傷造成的凋亡,其機(jī)制可能與增加抗氧化酶活性以及抑制Fas/Fas L蛋白表達(dá)來(lái)抑制細(xì)胞凋亡有關(guān),這為尋找緩解動(dòng)物熱應(yīng)激方法提供理論依據(jù)。
[Abstract]:Among the various physiological functions of mammals, reproductive function is the most vulnerable to heat stress. Heat stress causes oxidative stress in tissues, which destroys spermatogenesis in male animals, thus affecting reproductive efficiency. In order to investigate the effect of baicalin on testis injury of heat stress mice, and to explore the mechanism of anti-oxidation of baicalin, the mice were divided into 4 groups.
