【摘要】：為監(jiān)測(cè)和探究河南省豬流行性腹瀉病毒(porcine epidemic diarrhea virus,PEDV)流行株變異情況,本試驗(yàn)對(duì)15株2014-2015年間河南省PEDV流行株S基因進(jìn)行克隆及全序列測(cè)定,與河南省2011-2013年流行株以及國內(nèi)、外代表毒株進(jìn)行同源性及遺傳變異分析,并對(duì)這些毒株S基因的N-糖基化位點(diǎn)及跨膜螺旋區(qū)域進(jìn)行預(yù)測(cè)。結(jié)果表明:本試驗(yàn)得到的15株毒株之間S基因核苷酸同源性為97.0%~99.8%,氨基酸同源性為97.0%~99.5%;與2011-2013年河南流行株核苷酸同源性為97.1%~99.0%,氨基酸同源性為97.2%~98.8%,與其他地區(qū)流行毒株核苷酸同源性為92.2%~99.0%,氨基酸同源性為91.9%~99.0%;與經(jīng)典毒株相比,S1區(qū)域N端存在15bp的插入和6bp的缺失,核苷酸同源性為91.7%~93.6%,氨基酸同源性為92.0%~93.6%。系統(tǒng)發(fā)育建樹結(jié)果顯示目前的流行毒株與經(jīng)典毒株處于不同的2個(gè)群,流行毒株分處不同亞群。對(duì)N-糖基化位點(diǎn)及跨膜螺旋區(qū)域的預(yù)測(cè)得知2014-2015年河南省PEDV與經(jīng)典株相比出現(xiàn)變異,與2010年暴發(fā)時(shí)相比存在部分突變,毒株之間存在部分差異,研制出有效的針對(duì)流行毒的疫苗依然是目前防控該病的重中之重。
[Abstract]:In order to monitor and investigate the variation of S gene of porcine epidemic diarrhea virus in Henan Province, the S gene of 15 epidemic strains of PEDV in Henan Province from 2014 to 2015 were cloned and sequenced, and compared with those in Henan Province from 2011 to 2013 and in China. The homology and genetic variation of S gene were analyzed and the N-glycosylation sites and transmembrane helical regions of S gene were predicted. The results showed that the nucleotide homology of S gene and amino acid homology among the 15 strains were 97.0 ~ 99.8 and 97.0 ~ 99.5.The nucleotide homology and amino acid homology of the 15 strains were 97.1and 98.8 respectively, and the nucleotide homology and amino acid homology were 97.1and 98.8, respectively, and the nucleotide homology was 97.1%, and the amino acid homology was 98.8% with that of other endemic strains in Henan from 2011 to 2013. The homology of glycoside and amino acid was 92.2% and 91.9%, respectively. Compared with the classical strain, there was 15bp insertion and 6bp deletion in S _ 1 region. The homology of nucleotide and amino acid was 91.7 and 93.6, respectively. Phylogenetic results showed that the current epidemic strains and classical strains were in two different groups, and the epidemic strains had different subgroups. The prediction of N-glycosylation site and transmembrane helical region showed that the variation of PEDV in Henan Province in 2014-2015 was compared with that of classical strains, and there were some mutations and some differences among strains compared with the outbreak in 2010. The development of an effective vaccine against the epidemic is still a top priority in the prevention and control of the disease.
