和田羊KAPs、KRT候選基因的遺傳變異與羊毛細度和長度的相關性分析

【摘要】:本試驗運用PCR-SSCP和DNA測序技術,對和田羊角蛋白家族5個候選基因遺傳多態(tài)性進行了研究;利用SAS8.1最小二乘方差分析其與羊毛長度和細度的關系,并進一步分析了多基因組合對新疆和田羊羊毛性狀的影響。旨在找出與羊毛性狀相關基因,為今后和田羊羊毛性狀的改善以及育種工作提供科學依據(jù),從而提高牧民的經(jīng)濟收入。KAP6.1基因在堿基序列非編碼區(qū)內(nèi)含子發(fā)生C51T和G57A兩處突變;該基因與羊毛長度及細度關聯(lián)結果差異均不顯著(P0.05)。KAP7基因堿基在序列T110G和G268A處發(fā)生突變。AA和AB基因型的羊毛長度差異不顯著,AA、AB基因型與BB基因型差異極顯著;三個基因型對羊毛細度差異均不顯著。KAP7基因可能作為和田羊毛長度性狀的一個DNA分子標記。KRT27基因堿基序列中在G49A、T58C兩處發(fā)生突變;SS基因型與ST、TT基因型比較,羊毛長度及細度差異均顯著(0.01P0.05);ST基因型和TT基因型比較,羊毛長度及細度差異均極顯著(P0.01)。KRT27基因可能作為和田羊羊毛長度、細度的一個DNA分子標記。KRT31基因堿基序列發(fā)生突變C52T。與羊毛長度性狀關聯(lián)分析中,EE基因型和EF、FF基因型差異顯著(0.01P0.05),FF基因型極顯著長于EF基因型(P0.01);與羊毛細度性狀關聯(lián)分析中,三個基因型差異均不顯著(P0.05)。KRT31基因可能作為和田羊羊毛長度的一個DNA分子標記。本試驗五個基因中,KAP8基因未檢測出多態(tài)性;KAP6.1、KAP7、KRT27和KRT31在和田羊群體中突變位點均處于Hardy-Weinberg平衡狀態(tài)(P0.05)。在新疆和田羊群體中,利用基因聚合技術分析,組合合并基因型優(yōu)勢效應在羊毛細度上未見到體現(xiàn),而在羊毛長度上表現(xiàn)出了相應的優(yōu)勢。為今后進一步采用多基因聚合育種技術研究和田羊羊毛性狀提供了相應的參考依據(jù)。
[Abstract]:The genetic polymorphisms of five candidate genes of the Hetian sheep keratin family were studied by using PCR-SSCP and DNA sequencing techniques, and the relationship between the genetic polymorphisms of five candidate genes and wool length and fineness was analyzed by SAS8.1 least square variance analysis. The effects of polygene combinations on wool traits of Xinjiang Hetian sheep were further analyzed. The aim of this study was to find out the genes related to wool traits, and to provide scientific basis for the improvement of wool traits and breeding work in Hetian sheep in the future, so as to increase the economic income of herdsmen. KAP6.1 gene had two mutations in the intron of non-coding region of base sequence, C51T and G57A. There was no significant difference between the length of wool and the length and fineness of wool. There was no significant difference in wool length between AA and AB genotypes in sequence T110G and G268A, and there was no significant difference between AA AB genotype and BB genotype in sequence T110G and G268A. There was no significant difference in wool fineness between three genotypes. KAP7 gene might be used as a DNA molecular marker. KRT27 gene base sequence of the Hetian wool length trait had a mutation of SS genotype and STTTT genotype at two locations of G49AfT58C. There were significant differences in wool length and fineness between the St genotype and TT genotype of 0.01P0.05, and the differences of wool length and fineness were very significant. The gene P0.01U. KRT27 might be used as a DNA molecular marker for wool length of Hotan Sheep. The base sequence of the KRT31 gene was mutated C52T. In the correlation analysis with wool length traits, there were significant differences between EE genotype and EFF FF genotype (0.01P0.05F), which were significantly longer than that of EF genotype (P0.01A), and were correlated with wool fineness traits (P 0.01). The differences among the three genotypes were not significant (P 0.05N. KRT31). KRT31 gene might be used as a DNA molecular marker for wool length of Hetian sheep. In this study, no polymorphic KAP6.1, KAP7, KRT27 and KRT31 mutation loci were found in the Hardy-Weinberg equilibrium state of the five genes in the Hetian sheep population. In Xinjiang Hotan sheep population, the combination genotype dominance effect was not reflected in wool fineness, but in wool length. The results provided a reference basis for the further study of polygene polymerization breeding and wool traits in sheep.
