【摘要】:為探明大熊貓?jiān)词鄻有℃咦泳淖罴雅囵B(yǎng)條件,在不同培養(yǎng)基、碳源、氮源、溫度、pH等條件下培養(yǎng)石膏樣小孢子菌,用十字交叉法測(cè)量菌落每天的生長(zhǎng)情況。結(jié)果顯示,石膏樣小孢子菌在沙氏葡萄糖蛋白胨瓊脂培養(yǎng)基上生長(zhǎng)最好,在改良沙氏葡萄糖蛋白胨瓊脂培養(yǎng)基、馬鈴薯葡萄糖瓊脂培養(yǎng)基、高鹽培養(yǎng)基上均生長(zhǎng)良好,但在皮膚癬菌鑒定培養(yǎng)基和子囊孢子培養(yǎng)基上生長(zhǎng)緩慢;5種碳源對(duì)菌株生長(zhǎng)的影響不大,其中葡萄糖使該菌生長(zhǎng)較好;在5種氮源培養(yǎng)基中,除了在以尿素為氮源的培養(yǎng)基中該菌株不能生長(zhǎng)外,在以蛋白胨、硝酸鈉、草酸銨、硫酸銨為碳源的培養(yǎng)基中該菌均能生長(zhǎng),且在以蛋白胨為氮源的培養(yǎng)基中生長(zhǎng)最好;該菌株生長(zhǎng)的最適培養(yǎng)溫度為25℃,在15~35℃均可生長(zhǎng),在35℃時(shí)生長(zhǎng)最差;在pH值為10時(shí)生長(zhǎng)最好,在pH值為5、6、8、9時(shí)均能夠良好生長(zhǎng),比在pH值為7時(shí)的生長(zhǎng)更好,在pH值為4、11時(shí)的生長(zhǎng)較差。
[Abstract]:In order to find out the best culture conditions of Gypsum microspore from giant panda, microsporum gypsum was cultured in different culture medium, carbon source, nitrogen source, temperature and pH, and the growth of colony was measured by cross method. The results showed that the microsporozoa gypsum grew best on the salsa glucose peptone Agar medium, and on the modified Salvia glucose peptone Agar medium, the potato glucose Agar medium and the high salt medium. However, the effects of five carbon sources on the growth of Trichophyton dermatophytes and ascospore medium were not significant, and glucose made the strain grow better. Except that the strain could not grow in the medium with urea as nitrogen source, the strain could grow in the medium with peptone, sodium nitrate, ammonium oxalate and ammonium sulfate as carbon source, and the best growth was in the medium with peptone as nitrogen source. The optimum growth temperature of the strain is 25 鈩,
