利用纖維素降解菌固態(tài)發(fā)酵提高粗飼料營(yíng)養(yǎng)價(jià)值的研究

【摘要】:農(nóng)作物秸稈提供了豐富的粗飼料來(lái)源,但由于諸多營(yíng)養(yǎng)限制因素,不利于動(dòng)物的消化吸收。本論文利用綠色木霉、M-2菌株和熱帶假絲酵母發(fā)酵不同秸稈飼料,對(duì)其營(yíng)養(yǎng)成分變化進(jìn)行研究,并用體外產(chǎn)氣法對(duì)其營(yíng)養(yǎng)價(jià)值進(jìn)行評(píng)定,目的是研究不同菌種對(duì)秸稈營(yíng)養(yǎng)成分的影響。主要實(shí)驗(yàn)結(jié)果如下:(1)綠色木霉和M-2菌株單獨(dú)發(fā)酵六種不同秸稈飼料時(shí),都具有較高的纖維素酶活性。M-2菌株在玉米秸稈、甘蔗梢、木薯渣、象草培養(yǎng)基上的纖維素酶活性較綠色木霉高。六種秸稈飼料接種綠色木霉和M-2菌株發(fā)酵后粗蛋白含量都有顯著提高,半纖維素,纖維素都發(fā)生了一定的降解。(2)通過(guò)正交試驗(yàn),確定M-2菌株和熱帶假絲酵母在木薯渣培養(yǎng)基上的最優(yōu)發(fā)酵條件為:溫度35℃,料水比1:1.6,pH為7,接種M-2菌株60 h后接種熱帶假絲酵母。在此最佳發(fā)酵條件下,M-2菌株和熱帶假絲酵母混合發(fā)酵木薯渣后粗蛋白含量、半纖維素降解率、纖維素降解率分別為21.89%、58.36%和41.91%;M-2菌株和熱帶假絲酵母在玉米秸稈培養(yǎng)基上的最優(yōu)發(fā)酵條件為:溫度35℃,料水比1:1.4,pH為6,接種木霉84h后接種酵母。在此最佳發(fā)酵條件下,M-2菌株和熱帶假絲酵母混合發(fā)酵玉米秸稈后粗蛋白含量、半纖維素降解率、纖維素降解率分別為19.65%、35.70%和20.73%。(3)采用體外產(chǎn)氣法分別測(cè)定了不同發(fā)酵處理后秸稈飼料72h累計(jì)產(chǎn)氣量并與處理前進(jìn)行對(duì)照,結(jié)果表明,不同菌種發(fā)酵處理后秸稈飼料體外產(chǎn)氣量均有下降。由24h的累計(jì)產(chǎn)氣量計(jì)算得到各發(fā)酵秸稈飼料的有機(jī)物降解率、代謝能和消化能較對(duì)照組均有升高。
[Abstract]:Crop straw provides abundant forage sources, but it is not conducive to digestion and absorption of animals due to many nutritional constraints. In this paper, we used Trichoderma viride M-2 strain and Candida tropicalis to ferment different straw forage, and to study the change of nutrient composition and evaluate its nutritional value by in vitro gas production method. The aim of this study was to study the effect of different strains on the nutrient composition of straw. The main results were as follows: (1) Trichoderma viride and strain M-2 had higher cellulase activity in corn stalks, sugarcane shoots, cassava dregs. The cellulase activity in the medium was higher than that in Trichoderma viridis. After inoculation with Trichoderma viride and strain M-2, the crude protein content of six kinds of straw feed increased significantly, and hemicellulose and cellulose were all degraded.
