貴陽動物園動物隱孢子蟲的調查與蟲種鑒定

【摘要】:為了解貴陽動物園圈養(yǎng)野生動物隱孢子蟲的感染流行情況,以駱駝、黑熊、斑馬、袋鼠、獼猴、長臂猿、大象等野生動物的新鮮糞便為樣品(n=84),采用飽和蔗糖溶液漂浮法收集隱孢子蟲的卵囊并提取D N A,以18S r R NA為目標基因靶點設計特異性引物,并采用巢式PCR擴增目的基因,后經10 g/L瓊脂糖凝膠電泳,對檢測結果為陽性的PCR產物進行序列分析并建立系統(tǒng)發(fā)育樹。結果表明,共有13份隱孢子蟲陽性樣品,其中駱駝7份,感染率為70.00%,感染蟲種為安氏隱孢子蟲;斑馬1份,感染率為10.00%,感染蟲種為安氏隱孢子蟲;長臂猿5份,感染率為23.81%,感染蟲種為微小隱孢子蟲、人隱孢子蟲及小鼠隱孢子蟲。本次調查發(fā)現(xiàn)該園野生動物隱孢子蟲具有較高的總體感染率(15.50%)和鑒定出4種人獸共患隱孢子蟲蟲種,具有較重要的公共衛(wèi)生意義。本研究首次報道了我國斑馬感染隱孢子蟲。
[Abstract]:In order to understand the prevalence of cryptosporidium in captive wild animals in Guiyang Zoo, camels, black bears, zebras, kangaroos, macaques, gibbons, Fresh feces from wild animals such as elephants were used as samples. The oocysts of Cryptosporidium were collected by saturated sucrose solution floating method. The specific primers were designed with 18s r R NA as the target gene target, and the target genes were amplified by nested PCR. After 10 g / L agarose gel electrophoresis, the positive PCR products were sequenced and phylogenetic tree was established. The results showed that there were 13 positive samples of Cryptosporidium, of which 7 were camels, the infection rate was 70.00.The infection rate was Cryptosporidium andersoni, one zebra, the infection rate was 10.00, the infection rate was Cryptosporidium Angiosporium, and the gibbon was 5. The infection rate was 23.81%. The infected species were Cryptosporidium minimus, Cryptosporidium human and Cryptosporidium mouse. This investigation found that the wild animal Cryptosporidium has a higher total infection rate of 15.50%) and identified four species of zoonotic Cryptosporidium species, which has important public health significance. This study reported for the first time that zebras were infected with Cryptosporidium in China.
