【摘要】：為研究豬流感病毒(Swine influenza virus,SIV)A/swine/Dezhou/37/2015(H1N1)株的遺傳進(jìn)化和致病力,本研究對該分離株的8個(gè)基因節(jié)段進(jìn)行了遺傳進(jìn)化分析,并接種小鼠觀察臨床表現(xiàn)及檢測其肺臟的病理變化情況。結(jié)果顯示:分離株HA基因的裂解位點(diǎn)為PSIQSR↓G,無多個(gè)連續(xù)堿性氨基酸插入,為低致病性SIV;HA、M、NA節(jié)段處于歐洲類禽H1N1分支;NP、PA、PB1、PB2節(jié)段處于2009甲型H1N1分支;NS節(jié)段處于北美三源重排分支;對小鼠致病力研究顯示該毒株可感染小鼠并引起其肺臟組織病理學(xué)變化,但對小鼠無致死性。以上結(jié)果表明該分離株為一株低致病性的新型三源重排SIV。本研究為山東地區(qū)SIV的流行特點(diǎn)及變異情況提供了實(shí)驗(yàn)依據(jù)。
[Abstract]:In order to study the genetic evolution and pathogenicity of the Swine influenza virus (SIV) A/swine/Dezhou/37/2015 (H1N1) strain, the genetic evolution of 8 segments of the isolate was analyzed in this study. The clinical manifestations and pathological changes of the lungs were observed in mice. The results showed that the cleavage site of the HA gene of the isolated strain was found. For PSIQSR G, no successive alkaline amino acids were inserted into the low pathogenic SIV; HA, M, NA segments were in the European avian H1N1 branch; NP, PA, PB1, PB2 segments were in the 2009 type H1N1 branch; the segment was in the three source rearrangement branch of North America; the study of virulence in mice showed that the virus could infect mice and cause pathological changes of lung tissue, but against the pathological changes of lung tissue, but the The results showed that the isolated strain was a new type of low pathogenic three source rearrangement SIV., which provided experimental basis for the epidemic characteristics and variation of SIV in Shandong area.
