錳中毒導(dǎo)致雞肝臟炎癥相關(guān)因子mRNA表達(dá)增加以及肝臟損傷

【摘要】:錳,作為生物體內(nèi)必需的一種微量元素,廣泛分布在多種不同的生物體器官及組織內(nèi)。微量元素錳在機(jī)體的生長發(fā)育等方面都發(fā)揮著極其重要的作用。然而,金屬錳也屬于一種常見的環(huán)境污染物。當(dāng)生物體長時(shí)間處于在錳環(huán)境中,或者吸入過量錳的時(shí)候,不僅能夠?qū)C(jī)體產(chǎn)生不同程度的損傷,還能夠引起機(jī)體錳中毒,例如,錳中毒能夠?qū)ι窠?jīng)、消化、泌尿生殖以及免疫系統(tǒng)等正常生理功能造成一定程度的影響。目前,國內(nèi)外研究錳的毒性相關(guān)的實(shí)驗(yàn),多數(shù)都是以人和鼠作為研究對象展開的研究,而對于禽類錳毒性的研究和報(bào)道卻很少。所以,本試驗(yàn)以海藍(lán)雞為試驗(yàn)動物,我們的試驗(yàn)將180只1日齡健康的海藍(lán)雞隨機(jī)分成對照組、低劑量組、中劑量組和高劑量組共四組。對照組中飼喂正常日糧,而將低劑量組、中劑量組、高劑量組分別按錳對雞半數(shù)致死量的染毒量換算成飼料攝取量,進(jìn)行拌料染錳,使低劑量組、中劑量組、高劑量組中飼喂的日糧錳含量分別達(dá)到:600 mg/kg、900 mg/kg和1800mg/kg。在試驗(yàn)進(jìn)行的第30天、60天、90天每組隨機(jī)抽取15只雞,處死后取肝臟組織樣。試驗(yàn)內(nèi)容為觀察肝臟組織在光學(xué)顯微鏡和透射電子顯微鏡下的病理學(xué)變化,并檢測肝臟組織NO含量和i NOS活性,以及運(yùn)用實(shí)時(shí)定量PCR技術(shù)對雞肝臟組織中NF-κB、TNF-α、i NOS、COX-2和PGE2 m RNA表達(dá)量進(jìn)行檢測。研究錳過量對雞肝臟功能產(chǎn)生的影響,并初步探討錳中毒可能造成的毒理學(xué)機(jī)制。得出以下結(jié)果:(1)根據(jù)顯微觀察和超顯微觀察的結(jié)果,結(jié)果顯示雞肝臟組織發(fā)生損傷;(2)根據(jù)試劑盒測定結(jié)果發(fā)現(xiàn),雞肝臟組織中NO含量增加,i NOS活性升高;(3)實(shí)時(shí)定量PCR檢測的結(jié)果顯示出,雞肝臟組織中NF-κB、TNF-α、i NOS、COX-2和PGE2mRNA表達(dá)升高。我們的研究結(jié)果表明,過量的錳暴露能引起炎癥反應(yīng),使NF-κB、TNF-α、i NOS、COX-2和PGE2 m RNA表達(dá)增加,并且使雞肝臟組織中NO含量和i NOS活性提高。所得到的結(jié)果表明,雞肝臟中NF-κB途徑在錳誘導(dǎo)的炎癥反應(yīng)中起重要作用
[Abstract]:Manganese, as a necessary trace element in organisms, is widely distributed in many different organism organs and tissues. Trace element manganese plays an extremely important role in the growth and development of the body. However, manganese metal is also a common environmental pollutant. When an organism is in a manganese environment for a long time, or when it inhales excessive manganese, it can not only cause varying degrees of damage to the body, but also lead to manganese poisoning in the organism. For example, manganese poisoning can cause nerve, digestion, Normal physiological functions such as genitourinary and immune system are affected to a certain extent. At present, most of the experiments related to manganese toxicity at home and abroad are carried out with human and mouse as the research objects, but there are few studies and reports on poultry manganese toxicity. Therefore, 180 1-day-old healthy sea blue chickens were randomly divided into four groups: control group, low dose group, middle dose group and high dose group. The control group was fed normal diet, and the low dose group, middle dose group and high dose group were converted into feed intake according to the lethal dose of manganese to chicken, respectively, and mixed with manganese to dye manganese, so that the low dose group, the middle dose group, In the high dose group, the manganese content in the diet reached 900 mg/kg and 1800 mg / kg 路kg ~ (-1), respectively, at 1: 600 mg 路kg ~ (-1) 路kg ~ (-1) 路kg ~ (-1). 15 chickens were randomly selected from each group on the 30th day, 60 days and 90 days after the trial. To study the effect of manganese excess on liver function of chicken, and to explore the possible toxicological mechanism of manganese poisoning. According to the results of microscopic observation and ultramicroscopic observation, the result shows that the liver tissue of chicken is damaged. The results of real-time quantitative PCR detection showed that the expression of COX-2 and PGE2mRNA in chicken liver tissue was increased. Our results showed that excessive exposure to manganese could induce inflammatory reaction, increase the expression of COX-2 and PGE2 m RNA in NF- 魏 B, TNF- 偽 -TNF- 偽 I, and increase the content of no and the activity of I NOS in chicken liver. The results show that NF- 魏 B pathway plays an important role in manganese induced inflammation in chicken liver.
