ZEA、DON及其聯(lián)合染毒對(duì)CTLL-2細(xì)胞因子分泌的影響

【摘要】:為研究鐮刀菌毒素玉米赤霉烯酮(ZEA)、脫氧雪腐鐮刀菌烯醇(DON)及其聯(lián)合作用對(duì)動(dòng)物免疫功能的影響,試驗(yàn)以CTLL-2細(xì)胞(細(xì)胞毒性T淋巴細(xì)胞株)為材料,用不同濃度的ZEA(0、5、10、20μg/mL)、DON(0、0.5、1、2μg/mL)及聯(lián)合(空白組、5μg/mL ZEA、0.5μg/mL DON、5μg/mL ZEA+0.5μg/mL DON)處理CTLL-2細(xì)胞48h,采用ELISA法檢測(cè)了細(xì)胞內(nèi)及培養(yǎng)上清液中顆粒酶B(GZMB)、穿孔素(PFP)、干擾素γ(IFN-γ)和腫瘤壞死因子α(TNF-α)等細(xì)胞因子的含量。結(jié)果顯示,ZEA、DON能夠降低CTLL-2細(xì)胞胞內(nèi)及培養(yǎng)上清液中PFP、GZMB、IFN-γ的濃度,增加TNF-α濃度,染毒組與對(duì)照組相比均有顯著或極顯著差異(P0.05;P0.01),且均呈劑量效應(yīng)關(guān)系;ZEA、DON聯(lián)合染毒表現(xiàn)為加性效應(yīng)。結(jié)果表明,ZEA、DON及其聯(lián)合作用可通過影響免疫細(xì)胞因子的分泌,降低免疫細(xì)胞殺傷活力,間接影響機(jī)體體液免疫和細(xì)胞免疫的負(fù)調(diào)節(jié),從而導(dǎo)致動(dòng)物機(jī)體免疫機(jī)能下降。
[Abstract]:In order to study the effect of Fusoxin ZEAA, deoxynivalenoldol (DON) and its combined action on the immune function of animals, CTLL-2 cells (cytotoxic T lymphocyte strain) were used as materials. The cell line CTLL-2 cells were treated with different concentrations of ZEAA (0.5 渭 g/mL, 0.5 渭 g/mL ZEA, 0.5 渭 g/mL DON-1) and combined (5 渭 g/mL, 0.5 渭 g/mL DONs). The levels of cytokines such as granulase Bg/mL GZMBN, perforin PFPN, interferon 緯 IFN- 緯) and tumor necrosis factor- 偽 (TNF- 偽) were determined by ELISA assay. The results showed that ZEADON could decrease the concentration of IFN- 緯 in CTLL-2 cells and culture supernatant, and increase the concentration of TNF- 偽. Compared with the control group, there were significant or extremely significant differences between the two groups. The results showed that ZEADON and its combined action could decrease the killing activity of immune cells by affecting the secretion of immune cytokines and indirectly affect the negative regulation of humoral and cellular immunity, resulting in the decrease of immune function in animals.
