益陽(yáng)市犬弓首蛔蟲(chóng)感染情況調(diào)查及ITS、cox1基因的多態(tài)性分析

【摘要】:犬弓首蛔蟲(chóng)病是由犬弓首蛔蟲(chóng)(Toxocara canis)寄生于犬和其它動(dòng)物(包括人)而引起的一種動(dòng)物源性寄生蟲(chóng)病。目前湖南省益陽(yáng)市地區(qū)還沒(méi)有相關(guān)的研究報(bào)導(dǎo)。本文通過(guò)在2012年至2014年對(duì)湖南省益陽(yáng)市的犬弓首蛔蟲(chóng)的感染情況進(jìn)行調(diào)查,并運(yùn)用分子生物學(xué)方法對(duì)其核糖體內(nèi)部轉(zhuǎn)錄間隔區(qū)(ITS rDNA)和線粒體細(xì)胞色素氧化酶I基因(coxl)進(jìn)行多態(tài)性研究,得出結(jié)果如下:1.應(yīng)用飽和鹽水漂浮法檢查犬糞便樣品417份,其中229份檢測(cè)出犬弓首蛔蟲(chóng)蟲(chóng)卵,平均感染率為54.9%。2.驅(qū)蟲(chóng)試驗(yàn)結(jié)果顯示,芬苯達(dá)唑、伊維菌素、左旋咪唑三種藥物均能有效驅(qū)除犬弓首蛔蟲(chóng),其中芬苯達(dá)唑效果最佳。3.應(yīng)用分子生物學(xué)方法對(duì)犬弓首蛔蟲(chóng)的核糖體內(nèi)部轉(zhuǎn)錄間隔區(qū)(Internal transcrib ed spacer ribosome DNA -ITS rDNA)和犬弓首蛔蟲(chóng)的線粒體細(xì)胞色素氧化酶I基因(cytochrome c oxidase subunit 1-cox1)進(jìn)行多態(tài)性研究,結(jié)果顯示:11株犬弓首蛔蟲(chóng)均得到472 bp的ITS-1 rDNA序列,且種內(nèi)相似率為57.6%-76.9%,種間相似率為57.6%-76.9%;11株犬弓首蛔蟲(chóng)均得到323 bp的ITS-2 rDNA序列,序列種內(nèi)相似率達(dá)99.4%以上,種間相似率均在46.7%-77.4%之間;線粒體coxl基因的種內(nèi)相似性為99.3%-99.8%,種間相似性為85.8%-88.8%。
[Abstract]:Ascaris canis is an animal parasitic disease caused by Toxocara canis parasitic on dogs and other animals (including human beings). At present, there is no related research report in Yiyang area of Hunan Province. From 2012 to 2014, we investigated the infection of Ascaris canis in Yiyang City, Hunan Province. The polymorphism of its rDNA and mitochondrial cytochrome oxidase I gene was studied by molecular biology. The results were as follows: 1. The fecal samples of 417 dog feces were examined by saturated brine floating method, 229 of which were found to be eggs of Ascaris canis, and the average infection rate was 54.9. 2. The results of insect repellent test showed that fenbendazole, ivermectin and levamisole could effectively eliminate Ascaris canis, among which fenbendazole was the best. The polymorphism of internal transcrib ed spacer ribosome DNA-its rDNA in internal ribosomal spacer of Ascaris canis and cytochrome c oxidase subunit 1-cox1 gene of Ascaris canis was studied by molecular biology. The results showed that the ITS-1 rDNA sequence of 472bp was obtained in all 11 strains of Ascaris canis, and the intraspecific similarity rate was 57.6- 76.9. the interspecific similarity rate was 57.6- 76.9bp, and the ITS-2 rDNA sequence was 323bp for 11 strains of Ascaris canis. The similarity rate was 99.4% and 46.7- 77.4% respectively. The intraspecific similarity of mitochondrial coxl gene was 99.3-99.8, and the interspecific similarity was 85.8% -88.8%.
