山羊CCRL2基因的分子克隆和表達分析

【摘要】:本研究旨在克隆山羊趨化因子(C-C基序)受體2(chemokine(C C motif)receptor-like 2,CCRL2)基因序列,并研究其組織和時序表達譜。以簡州大耳羊為試驗材料,應(yīng)用RT-PCR方法克隆CCRL2基因并進行生物信息學分析,qPCR方法構(gòu)建該基因在山羊不同組織表達譜和不同月齡背最長肌、股二頭肌和臂三頭肌中的表達水平,并將基因表達與IMF含量進行相關(guān)分析。結(jié)果顯示:克隆得到山羊CCRL2基因(GenBank登錄號:KT165378)長度為1 143bp,編碼區(qū)長度為1 047bp;qPCR分析表明,CCRL2mRNA在24月齡山羊心、肝、脾、肺、腎、脂肪、背最長肌、股二頭肌和臂三頭肌中均有不同程度的表達,其中脾表達量最高(P0.01);時序表達中,CCRL2基因在股二頭肌和臂三頭肌中的表達量均在8~10月齡極顯著高于1~3和24月齡(P0.01),在背最長肌中則為24月齡極顯著高于1~3和8~10月齡(P0.01);相關(guān)分析顯示,股二頭肌中CCRL2基因mRNA表達量與IMF含量呈極顯著負相關(guān)(P0.01)。CCRL2基因可能參與山羊IMF沉積作用。
[Abstract]:The aim of this study was to clone the gene sequence of goat chemokine C-C motif) receptor 2(chemokine(C C motif)receptor-like 2and CCRL2, and to study its tissue and temporal expression profile. The CCRL2 gene was cloned by RT-PCR and analyzed by bioinformatics to construct the expression profile of the gene in different tissues of goat and the expression level of longissimus dorsi muscle, biceps femoris muscle and triceps arm muscle at different months of age. The correlation between gene expression and IMF content was analyzed. The results showed that the length of goat CCRL2 gene was 1143 BP and the length of coding region was 1 047 BP / qPCR. The results showed that CCRL2 mRNA was found in 24 months old goat heart, liver, spleen, lung, kidney, fat and longissimus dorsi muscle. Both biceps femoris and triceps brachii showed different degrees of expression. The expression of CCRL2 gene in biceps femoris and triceps brachii muscle was significantly higher than that in the 1st and 24th months of age, and in the longissimus dorsi muscle, the expression of CCRL2 gene was significantly higher in the longissimus dorsi muscle than that in the 1st and 10th months of age, and the correlation analysis showed that the expression of CCRL2 gene in the biceps femoris and triceps brachii muscle was significantly higher than that in the longissimus dorsi muscle. The mRNA expression of CCRL2 gene in biceps femoris was negatively correlated with the content of IMF. CCRL2 gene may be involved in the deposition of IMF in goat.
