妊娠后期宮內(nèi)生長(zhǎng)限制對(duì)蒙古綿羊胎兒肝臟細(xì)胞增殖及其相關(guān)基因表達(dá)的影響

【摘要】:本論文系統(tǒng)的研究了妊娠后期宮內(nèi)生長(zhǎng)限制對(duì)蒙古綿羊胎兒肝臟細(xì)胞增殖及相關(guān)基因表達(dá)的影響。選取健康蒙古綿羊(經(jīng)同期發(fā)情受孕)24只。妊娠90 d時(shí),選擇與總體平均體重接近的6只母羊進(jìn)行屠宰,剩余綿羊按體重隨機(jī)分配到3個(gè)營(yíng)養(yǎng)水平處理組:0.18MJME·BW-0.75·d-1 (RG1:n=6)、0.33 MJME·BW-0.75·d-1 (RG2:n=6)和自由采食(CG:n=6),按以上營(yíng)養(yǎng)水平飼養(yǎng)至妊娠140 d,將每組6只母羊進(jìn)行屠宰實(shí)驗(yàn)。試驗(yàn)結(jié)果表明:(1)妊娠后期限飼母羊?qū)е孪揎暯M胎兒RG2 (p0.05),RG1組(p0.01)胎兒肝臟重,RG1組胎兒肝臟中ⅠCol (p<0.01)、 ⅢCol (p0.05)、ⅣCol(p<0.01) LN (p0.01), HA (p0.05)以及FN(p0.01),RG2組胎兒肝臟FN(p0.01)均顯著低于CG組。RG2、RG1組胎兒肝臟組織結(jié)構(gòu)受到嚴(yán)重影響,網(wǎng)狀纖維和膠原纖維明顯增多變粗。(2) RG2、RG1組胎兒肝臟DNA濃度值、蛋白質(zhì)/DNA均顯著低于CG組(p0.01); RG1組G0/G1期細(xì)胞百分比顯著高于RG2組(p0.05)、CG組(p0.01);RG2組(p0.05)、RG1組(p0.01)S期細(xì)胞百分比均顯著低于CG組:且RG2組(p0.05)、RG1組(p0.01)增殖指數(shù)均顯著低于CG組。(3)RG1組、RG2組CyclinA蛋白的表達(dá)量顯著低于CG組(p0.05),且RG1組、RG2組CyclinB,,CyclinE蛋白的表達(dá)量也顯著低于CG組(p0.01),RGl組CyclinD蛋白的表達(dá)量顯著高于RG2組(p<0.05); RG1、RG2組CDK1蛋白表達(dá)量(p<0.05)、CDK2蛋白表達(dá)量(p<0.01)、 CDK4蛋白表達(dá)量(p0.01)顯著低于CG組;RG1組E2F2蛋白表達(dá)量顯著低于RG2組、CG組(p0.01);RG1組E2F5蛋白表達(dá)量顯著高于RG2組和CG組(p<0.01); CG、RG2、RGl組胎兒肝臟中CDK1、 CyclinD1、E2F5基因表達(dá)量逐漸上調(diào),而CyclinB1、CDK4、E2F1基因表達(dá)量逐漸下調(diào)。
[Abstract]:The effects of intrauterine growth restriction on the proliferation of fetal liver cells and the expression of related genes in Mongolian sheep were systematically studied in this paper. A total of 24 healthy Mongolian sheep were selected. At the 90th day of gestation, 6 ewes, which were close to the total average body weight, were slaughtered. The remaining sheep were randomly divided into 3 groups according to their body weight: 0. 18MJME BW-0.75 d-1 RG1: nongfong 0.33 MJME BW-0.75 d-1 RG2: nongf6) and free feed CG2: nongf6). Six ewes of each group were slaughtered according to the above nutrition level until gestation 140d.
