牦牛PPARδ基因多態(tài)性與生長(zhǎng)性狀的相關(guān)性

【摘要】:【目的】揭示牦牛PPARδ基因的遺傳變異特征,發(fā)現(xiàn)與牦牛生長(zhǎng)性狀顯著相關(guān)的候選分子標(biāo)記,為牦牛分子育種積累基礎(chǔ)資料.【方法】以96頭牦牛血樣為材料,運(yùn)用DNA池測(cè)序和PCR-HRM等技術(shù)檢測(cè)PPARδ基因的序列變異.【結(jié)果】檢測(cè)到牦牛PPARδ基因內(nèi)含子中2個(gè)SNPs.遺傳變異特征分析顯示,SNP1(g.10045A/G)和SNP2(g.10226A/G)位點(diǎn)屬于中度多態(tài)性.連鎖不平衡和單倍型分析表明,牦牛PPARδ基因2個(gè)多態(tài)位點(diǎn)之間為弱連鎖,單倍型AA屬于優(yōu)勢(shì)單倍型(頻率為42.2%).方差分析表明,單個(gè)的SNP與牦牛生長(zhǎng)性狀有著顯著或極顯著的關(guān)聯(lián).【結(jié)論】牦牛PPARδ基因的2個(gè)SNPs與其生長(zhǎng)性狀顯著相關(guān),此結(jié)果可以應(yīng)用于牦牛的分子育種實(shí)踐.
[Abstract]:[objective] to reveal the genetic variation characteristics of yak PPAR 未 gene, and to find candidate molecular markers significantly related to yak growth traits, and to accumulate basic data for yak molecular breeding. [methods] 96 yak blood samples were used as materials. DNA pool sequencing and PCR-HRM were used to detect the sequence variation of PPAR 未 gene. [results] two SNPs in PPAR 未 gene intron of yak were detected. Genetic variation analysis showed that the SNP1 / 10045A / G) and SNP2 / 10226A / G) loci were moderately polymorphic. Linkage disequilibrium and haplotype analysis showed that the two polymorphic loci of yak PPAR 未 gene were weakly linked, and haplotype AA belonged to the dominant haplotype (the frequency was 42.2%). The analysis of variance showed that there was a significant or extremely significant correlation between single SNP and yak growth traits. [conclusion] two SNPs of yak PPAR 未 gene were significantly correlated with their growth traits, which could be applied to the practice of yak molecular breeding.
