豬圓環(huán)病毒3型PCR診斷方法的建立

【摘要】:根據(jù)NCBI中PCV3的基因組序列,設(shè)計(jì)了1對特異性引物,并對PCR的反應(yīng)體系和反應(yīng)條件進(jìn)行優(yōu)化,同時(shí)進(jìn)行特異性和敏感性試驗(yàn),最后應(yīng)用所建立的PCR方法檢測了臨床送檢的22份病料。結(jié)果顯示,建立的PCR方法的最佳反應(yīng)條件為總體積25 L,退火溫度56℃,模板DNA量3 L(l ng/m L),進(jìn)行35個(gè)循環(huán)。該P(yáng)CR能特異地?cái)U(kuò)增出目的片段,長度約為381 bp。測序結(jié)果表明,PCR擴(kuò)增產(chǎn)物正確。該P(yáng)CR反應(yīng)特異性強(qiáng),只能擴(kuò)增PCV3陽性樣品,且能擴(kuò)增的最低模板量為1×10-4ng/m L。用該P(yáng)CR對送檢的22份病料進(jìn)行PCV3檢測,結(jié)果陽性率為13.6%?梢,本研究建立的PCR方法特異、敏感,可用于PCV3的快速檢測。
[Abstract]:According to the genomic sequence of PCV3 in NCBI, a pair of specific primers was designed, and the reaction system and reaction conditions of PCR were optimized, and the specificity and sensitivity tests were carried out.
