【摘要】:目的:動(dòng)物毛纖維是人類生產(chǎn)生活不可或缺的重要資源,主要毛用性狀有細(xì)度、長(zhǎng)度和色彩。而毛色作為主要毛用性狀之一,是由許多基因共同決定的。老鼠作為重要的實(shí)驗(yàn)動(dòng)物,對(duì)其毛色的研究有助于對(duì)于其他動(dòng)物毛色的研究。方法:為了研究Stx17基因與小鼠毛色形成中的相關(guān)性,本試驗(yàn)通過在小鼠皮膚組織和小鼠的黑色素細(xì)胞上擴(kuò)增的Stx17基因的序列,并用熒光定量實(shí)時(shí)PCR、免疫化學(xué)和Western blot對(duì)其在不同毛色皮膚組織中進(jìn)行定量和定性的分析,以說明Stx17基因在不同毛色的小鼠的皮膚組織上的表達(dá)差異,從而為進(jìn)一步研究Stx17基因在不同毛色形成過程中的作用機(jī)制打好基礎(chǔ)。結(jié)果:(1)通過PCR技術(shù)成功擴(kuò)增出Stx17基因全cds區(qū)序列,經(jīng)測(cè)序所得為目的片段,在NCBI上與其他物種進(jìn)行同源序列的比對(duì),得知與其他物種的基因有較高的同源性。(2)通過Real-Time PCR研究stx17基因在不同毛色小鼠皮膚組織中的相對(duì)表達(dá)量,結(jié)果顯示白鼠和灰腹表達(dá)水平分別是黑背的0.115倍和1.682倍,差異極顯著(P0.01),可見野生型小鼠的灰色腹部中的基因表達(dá)量顯著高于白色小鼠個(gè)體和黑色背部,這說明該基因與灰色的形成有一定的相關(guān)性。(3)應(yīng)用免疫印跡技術(shù)(Western Blotting)對(duì)STX17蛋白在不同毛色皮膚組織中的蛋白表達(dá)進(jìn)行研究,結(jié)果顯示在白色小鼠、灰色和黑色小鼠皮膚組織中均有蛋白的表達(dá),并且在灰色小鼠皮膚組織中的表達(dá)量是最高的。(4)通過免疫化學(xué)法觀察Stx17基因在小鼠皮膚組織中的定位,結(jié)果顯示Stx17基因在小鼠皮膚的真皮部位呈陽性表達(dá)。并對(duì)小鼠的黑色素細(xì)胞進(jìn)行免疫化學(xué)染色后也證實(shí)STX17在黑素細(xì)胞的細(xì)胞膜上有表達(dá)。結(jié)論:以上所述表明,Stx17基因在不同毛色的皮膚組織中都有表達(dá),而且表達(dá)量有一定的差異性,因此,推測(cè)該基因可能與不同毛色的形成具有相關(guān)性。
[Abstract]:Objective: animal hair fiber is an indispensable resource for human production and life. The main wool characters are fineness, length and color. As one of the main wool traits, coat color is determined by many genes. As an important laboratory animal, the study of the hair color of mice is helpful to the study of other animal's coat color. Methods: in order to study the correlation between Stx17 gene and mouse coat color formation, the Stx17 gene sequence was amplified from mouse skin and melanocytes. Fluorescence quantitative real-time PCR, immunocytochemistry and Western blot were used to quantitatively and qualitatively analyze the expression of Stx17 gene in the skin tissues of mice with different coat colors, so as to explain the difference in the expression of Stx17 gene in the skin tissues of mice with different coat colors. The results provide a basis for further study on the mechanism of Stx17 gene in the process of different coat color formation. Results the whole cds region of Stx17 gene was successfully amplified by PCR. The sequence was sequenced and compared with other species on NCBI. The results showed that the relative expression of stx17 gene in the skin of different hairy mice was 0.115 times and 1.682 times higher than that of black back, respectively, and the expression level of stx17 gene was 0.115 times and 1.682 times higher than that of black back, respectively, by using Real-Time PCR, the results showed that the expression level of stx17 gene was 0.115 times and 1.682 times higher than that of black back, respectively. The difference was very significant (P 0.01). It was found that the gene expression in gray abdomen of wild type mice was significantly higher than that of white mice and black back. The results showed that the expression of STX17 protein in different skin tissues was studied by Western blotting technique. The results showed that the expression of STX17 protein was found in white mice. The expression of protein was found in both gray and black mouse skin tissues, and the highest expression level was found in gray mouse skin tissues.) Immunochemical method was used to observe the localization of Stx17 gene in mouse skin tissues. The results showed that Stx17 gene was positively expressed in the dermis of mice. Immunocytochemical staining of mouse melanocytes also confirmed that STX17 was expressed on melanocyte membrane. Conclusion: these results suggest that the Stx17 gene is expressed in skin tissues with different coat colors, and the expression levels are different. Therefore, it is speculated that this gene may be related to the formation of different coat colors.
