低溫脫脂大豆粕的低豐度蛋白提取和高豐度蛋白去除條件的優(yōu)化(英文)

【摘要】:目的:優(yōu)化低溫脫脂大豆粕中的低豐度蛋白提取和高豐度蛋白去除條件,為進(jìn)一步探討大豆低豐度蛋白對動物的生理功能影響提供試驗材料。創(chuàng)新點:結(jié)合提取時間和異丙醇濃度,將超聲波(超聲時間、功率)用于輔助異丙醇去除大豆高豐度蛋白和富集低豐度蛋白的研究。方法:在單因素試驗基礎(chǔ)上,設(shè)計了超聲時間、超聲功率、提取時間和異丙醇濃度等四因素三水平的正交試驗。并通過測定提取液中的蛋白濃度和通過聚丙烯酰胺凝膠電泳(SDS-PAGE)、質(zhì)譜分析(MS)等蛋白質(zhì)組學(xué)手段鑒定提取液或丙酮沉淀物中的蛋白質(zhì)含量,實現(xiàn)了大豆高豐度蛋白去除和低豐度蛋白提取條件的優(yōu)化。結(jié)論:50%異丙醇,350 W超聲15 min,提取1 h是大豆低豐度蛋白提取的最佳條件。提取物含多種低豐度蛋白(油質(zhì)蛋白、大豆未知蛋白、磷酸酯D和無特征大豆蛋白),且無大豆高豐度蛋白(大豆球蛋白和β-伴大豆球蛋白)。
[Abstract]:Objective: to optimize the conditions of low abundance protein extraction and high abundance protein removal from low temperature defatted soybean meal, and to provide experimental materials for further study on the effect of soybean low abundance protein on physiological function of animals. Innovation: combined with extraction time and isopropanol concentration, ultrasonic wave (ultrasonic time, power) was used to assist isopropanol to remove high abundance soybean protein and enrich low abundance protein. Methods: on the basis of single factor experiment, orthogonal experiments were designed with four factors: ultrasonic time, ultrasonic power, extraction time and isopropanol concentration. The protein concentration in the extract was determined and the protein content in the extract or acetone precipitate was identified by means of proteomics, such as polyacrylamide gel electrophoresis, SDS-PAGEG, MS, etc. The extraction conditions of soybean high abundance protein and low abundance protein were optimized. Conclusion the optimum extraction condition of soybean protein with low abundance was 50% isopropanol 350W for 15 min and 1 hour for extraction. The extract contains a variety of low abundance proteins (oil protein, soybean unknown protein, phosphate D and non-characteristic soybean protein), and no soybean high abundance protein (soybean globulin and 尾 -concomitant soybean globulin).
