IFNAR1和IFNAR2在妊娠早期牛胎盤組織中的表達研究

【摘要】:干擾素-τ(即IFNT)是一種I型干擾素,它是一種由妊娠滋養(yǎng)層分泌的抗黃體溶解因子。IFNT通過結合其受體IFNAR1和IFNAR2傳遞信號或者刺激某些因子的表達以建立母體妊娠識別信號。截止目前,有關IFNT受體(IFNAR1和IFNAR2)在奶牛胎盤組織上的表達定位及其作用的研究尚不夠完善。本研究運用石蠟切片制作、HE及PAS染色技術對妊娠早期牛胎盤顯微結構進行觀察,以確定牛胎盤子葉及肉阜結構特征及發(fā)育特點,形成妊娠早期牛胎盤發(fā)育的組織結構圖譜;進一步通過免疫熒光染色和qRT-PCR技術對妊娠早期牛胎盤組織中IFNAR1、IFNAR蛋白及其mRNA的細胞定位和表達規(guī)律進行研究,分析IFNAR1和IFNAR在妊娠早期牛胎盤發(fā)育中的功能意義。結果如下:1.妊娠早期的牛胎盤為子葉型/上皮絨毛膜胎盤,其子宮肉阜為半圓的穹頂狀。隨著孕齡增大,胎兒尿囊絨毛膜絨毛不斷分枝、變長、并增大與母體子宮內膜上皮的嵌合,從而不斷加大與母體胎盤區(qū)子宮內膜上皮的聯(lián)系。妊娠27d后,胎兒絨毛膜上皮由單核細胞和PAS染色各異的三類滋養(yǎng)層雙核巨細胞組成,并存在雙核巨細胞遷移進入肉阜陷窩與子宮內膜上皮細胞融合而成的合胞體現(xiàn)象;妊娠50d,可觀察到程度較淺的胎盤血腫。2.免疫熒光染色結果表明,在各日齡牛胎盤的尿囊絨毛膜、胎兒滋養(yǎng)層及母體子宮內膜均檢測到IFNAR1的表達,陽性反應主要出現(xiàn)于尿囊絨毛膜上皮、子宮內膜上皮、子葉、母體子宮固有層內子宮腺及腺間結締組織,其中IFNAR1在尿囊絨毛膜上皮、子宮內膜上皮及子宮腺呈強表達。免疫熒光相對表達量分析表明,IFNAR1在牛胎盤組織各部位的表達均隨妊娠日齡增大而增強。3.qRT-PCR結果表明,在妊娠早期的尿囊絨毛膜和子葉組織,IFNAR1和IFNAR2 mRNA的表達隨孕齡增大而不斷增加,且IFNAR1 mRNA的表達水平整體上要高于IFNAR2的表達。結論:在奶牛妊娠早期,胎兒胎盤通過不斷分枝、變長的胎兒尿囊絨毛膜絨毛增大與母體胎盤的子宮內膜上皮的嵌合而不斷加大母胎之間的聯(lián)系;妊娠早期牛胎盤組織中IFNAR1蛋白及IFNAR1和IFNAR2 mRANA呈規(guī)律性表達,提示兩種受體在牛的早期妊娠維持和胎兒發(fā)育中發(fā)揮重要作用。研究結果不僅豐富了反芻動物胎盤組織形態(tài)學理論,也為胎盤滋養(yǎng)層細胞的相關基礎和應用研究提供了基礎形態(tài)資料,并為闡明IFNAR1和IFNAR在牛胎盤發(fā)育過程中的作用機制提供理論依據(jù)。
[Abstract]:Interferon - tau (IFNT) is a type I interferon, which is an anti luteinolytic factor.IFNT secreted by the gestational trophoblastic layer by combining its receptor IFNAR1 and IFNAR2 transmission signal or stimulating some factors to establish a maternal pregnancy recognition signal. At present, the IFNT receptor (IFNAR1 and IFNAR2) is present in the placental tissue of dairy cows. The study of expression and its role is not perfect. In this study, the microstructure of bovine placenta in early pregnancy was observed by paraffin section, HE and PAS staining techniques to determine the structural characteristics and development characteristics of the plate and caruncle of the fetus and to form the tissue structure of the bovine placenta in the early pregnancy; further through immunofluorescence staining The location and expression of IFNAR1, IFNAR protein and mRNA in bovine placenta in early pregnancy were studied by color and qRT-PCR techniques. The functional significance of IFNAR1 and IFNAR in the development of bovine placenta in early pregnancy was analyzed. The results were as follows: 1. the bovine placenta in the early pregnancy was cotyledon / upper chorion placenta and the caruncle of the uterus was a semicircular dome. With the increase of gestational age, the chorionic villus of the fetal allantoin is continuously branched, longer, and increased with the endometrium epithelium of the mother body, thus increasing the relationship with the endometrium epithelium in the maternal placenta area. After 27D, the fetal chorionic epithelium consists of mononuclear cells and PAS dyed three types of mononuclear cells with different types of trophoblast. The migration of nuclear giant cell into the syncytial body formed by the fusion of caruncle lacuna and endometrium epithelial cells; pregnancy 50D, the results of.2. immunofluorescence staining of shallow placental hematoma showed that the expression of IFNAR1 was detected in the urinal chorionic villus, fetal trophoblastic and maternal endometrium at every day of age, and the positive reaction was main. In the urinal chorionic epithelium, the endometrium, the cotyledon, the endometrium and the connective tissue in the matrix of the mother's uterus, IFNAR1 is in the urinal chorionic epithelium, the endometrium epithelium and the uterus are strongly expressed. The expression of the immunofluorescence relative expression indicates that the expression of IFNAR1 in the various parts of the bovine placenta is increased with the age of pregnancy. The results of enhanced.3.qRT-PCR showed that the expression of IFNAR1 and IFNAR2 mRNA in the chorionic chorionic and cotyledon tissues of the early pregnancy increased with the increase of gestational age, and the expression level of IFNAR1 mRNA was higher than the expression of IFNAR2 on the whole. Conclusion: in the early pregnancy of the dairy cows, the fetal placenta is continuously branched and the fetal chorionic cashmere is longer. The relationship between Mao Zengda and the endometrium epithelium of the maternal placenta increases the relationship between the mother fetus and the mother fetus; the IFNAR1 protein and IFNAR1 and IFNAR2 mRANA in the bovine placenta tissue are regularly expressed in the early pregnancy, suggesting that the two receptors play an important role in the maintenance of early pregnancy and the development of the fetus. The results not only enrich the ruminant animals, but also enrich the ruminant. The theory of placental histomorphology also provides basic morphological data for the related basis and application of placental trophoblast cells, and provides a theoretical basis for elucidating the mechanism of IFNAR1 and IFNAR in the development of bovine placenta.
