烏珠穆沁羊骨骼肌肌纖維組織學(xué)特性及MyHC表達(dá)量變化分析

【摘要】:本實(shí)驗(yàn)采用6月齡、9月齡、12月齡與18月齡烏珠穆沁羊?yàn)檠芯繉ο?屠宰后取半腱肌、背最長肌、肱三頭肌和股二頭肌。利用ATPa.se組織化學(xué)染色法進(jìn)行肌纖維染色,探索不同月齡不同部位骨骼肌的肌纖維密度、類型的百分比、面積比例及直徑等組織學(xué)特性的變化;測定不同月齡烏珠穆沁羊各部位骨骼肌中肌球蛋白重鏈相關(guān)基因的表達(dá)水平;檢測各樣品硬度、咀嚼度及恢復(fù)性等品質(zhì)指標(biāo)。結(jié)果顯示:隨著月齡的增加各種肌纖維的直徑及橫截面積隨之增加,而單位面積肌纖維的數(shù)量會減少。從肌肉組織中不同類型肌纖維的百分比來看,Ⅱb型肌纖維百分比較多、其次為Ⅱa型肌纖維和Ⅰ型肌纖維。整體上隨著月齡的增加Ⅱb型肌纖維含量緩慢的增多,尤其背最長肌中Ⅱb型肌纖維的變化較為明顯;虮磉_(dá)結(jié)果顯示MyHC Ⅰ表達(dá)量在6月齡到9月齡有稍微的增長,在12月齡和18月齡都有所降低。MyHC Ⅱa表達(dá)量在9月齡時變化不顯著,12月齡開始降低,18月齡時達(dá)到最低值。MyHCⅡb和MyHCⅡx的表達(dá)量隨著月齡的增加顯著增加。相同月齡不同部位之間相比背最長肌MyHCⅡb表達(dá)量明顯大于其他三個部位。從質(zhì)構(gòu)儀檢測到的數(shù)據(jù)能看出烏珠穆沁羊隨著月齡的增加肉質(zhì)變得較硬,咀嚼度提高。因此月齡越大肌肉的嫩度越差。在四個不同部位之間背最長肌的變化最為明顯。相關(guān)性數(shù)據(jù)顯示肉的硬度和咀嚼度與MyHC Ⅰ和MyHC Ⅱ呈負(fù)相關(guān)性、與MyHC Ⅱx和MyHCⅡb呈正相關(guān)性、還與肌纖維密度呈負(fù)相關(guān)性,與肌纖維直徑呈正相關(guān)性。
[Abstract]:In this experiment, Uzhimuqin sheep (6 months old, 9 months old, 12 months old and 18 months old) were used as research objects. After slaughtering, the semitendinosus muscle, the longest dorsal muscle, triceps brachii muscle and biceps femoris muscle were taken. ATPa.se histochemical staining was used to study the changes of muscle fiber density, percentage of type, area ratio and diameter of skeletal muscle at different months of age. The expression levels of myosin heavy chain related genes in skeletal muscle of different month old Wudhumqin sheep were determined and the quality indexes such as hardness chewing degree and recovery of each sample were detected. The results showed that the diameter and cross-sectional area of muscle fibers increased with the increase of age, but the number of muscle fibers per unit area decreased.
