脂筏在牛呼吸道合胞體病毒感染MDBK細胞中的作用

【摘要】:為探討脂筏在牛呼吸道合胞體病毒(bovine respiratory syncytial virus,BRSV)感染MDBK細胞中的作用,采用膽固醇去除藥物MβCD處理MDBK細胞。結(jié)果顯示,細胞膜膽固醇含量明顯降低,同時也降低BRSV感染水平,補充外源膽固醇后,病毒感染力可部分恢復(fù);表明細胞膜膽固醇的去除可以抑制BRSV的感染,BRSV的感染與細胞膜脂筏相關(guān)。進一步研究發(fā)現(xiàn),膽固醇去除不會影響B(tài)RSV結(jié)合在靶細胞表面,但會抑制病毒進入靶細胞。在病毒感染后采用膽固醇去除藥物MβCD處理MDBK細胞,對于病毒感染力的影響是輕微的。這些發(fā)現(xiàn)可有助于理解BRSV的感染機制,為研制安全、高效的疫苗和有效的治療藥物提供理論依據(jù)。
[Abstract]:To investigate the role of lipid rafts in the infection of bovine respiratory syncytial virus (BRSVV) with bovine respiratory syncytial virus, MDBK cells were treated with cholesterol removal drug M 尾 CD.The results showed that the content of plasma membrane cholesterol was significantly decreased and the level of BRSV infection was also decreased. After supplementation of exogenous cholesterol, the viral infectivity could be partially recovered.The results showed that the removal of membrane cholesterol could inhibit the infection of BRSV.Further studies showed that cholesterol removal did not affect the binding of BRSV to target cells, but inhibited virus entry into target cells.After virus infection, MDBK cells were treated with cholesterol removal drug M 尾 CD, and the effect on viral infectivity was slight.These findings may be helpful to understand the mechanism of BRSV infection and provide theoretical basis for the development of safe, efficient vaccines and effective therapeutic drugs.
