【摘要】：本試驗(yàn)旨在研究飼糧中添加沙蔥黃酮對(duì)肉羊外周血淋巴細(xì)胞轉(zhuǎn)化率和凋亡率的影響。選用60只6月齡左右、體重(39.9±3.2)kg的小尾寒羊?yàn)樵囼?yàn)動(dòng)物,按照出生月齡和體重相近的原則,隨機(jī)分為4組,每組15只羊。對(duì)照組飼喂基礎(chǔ)飼糧,3個(gè)試驗(yàn)組分別在基礎(chǔ)飼糧中添加11、22、33 mg/kg的沙蔥總黃酮。預(yù)試期為15 d,正試期為60 d。正試期第60天空腹頸靜脈采血,采用噻唑藍(lán)(MTT)法測(cè)定肉羊外周血淋巴細(xì)胞轉(zhuǎn)化率,采用流式細(xì)胞儀測(cè)定細(xì)胞周期、細(xì)胞凋亡率。結(jié)果表明:1)飼糧添加11、22、33 mg/kg沙蔥黃酮可提高肉羊外周血淋巴細(xì)胞轉(zhuǎn)化率,其中33 mg/kg組顯著高于對(duì)照組(P0.05);2)與對(duì)照組相比,飼糧添加33 mg/kg沙蔥黃酮能夠顯著提高S期和G2/M期細(xì)胞比例(P0.05);3)與對(duì)照組和11 mg/kg組相比,22和33 mg/kg組外周血淋巴細(xì)胞凋亡率顯著提高(P0.05)。由此可見,飼糧中添加11~33 mg/kg沙蔥黃酮可促進(jìn)肉羊外周血淋巴細(xì)胞轉(zhuǎn)化,并使細(xì)胞向DNA合成期轉(zhuǎn)化,促進(jìn)細(xì)胞分裂,對(duì)細(xì)胞凋亡有顯著促進(jìn)作用,以添加33 mg/kg沙蔥黃酮的效果最佳。
[Abstract]:The purpose of this study was to study the effects of flavanone on lymphocyte transformation and apoptosis in peripheral blood of meat sheep.60 small tail Han sheep of about 6 months old and 39.9 鹵3.2)kg weight were selected as experimental animals. According to the principle of similar birth age and body weight, 60 small tail Han sheep were randomly divided into 4 groups with 15 sheep in each group.The control group was fed with basic diet, and the three experimental groups were fed with total flavonoids of Allium mongolicum for 11 ~ 22 ~ 33 mg/kg respectively.The pre-test period was 15 days and the positive test period was 60 days.Fasting jugular vein blood was collected on the 60th day of the trial period. The lymphocyte transformation rate in peripheral blood of sheep was measured by thiazolyl methoate (MTT) method. The cell cycle and apoptosis rate were measured by flow cytometry (FCM).The results showed that the lymphocyte transformation rate in peripheral blood of meat sheep was significantly higher in 33 mg/kg group than that in control group (P 0.05).Compared with control group and control group and 11 mg/kg group, the apoptosis rate of peripheral blood lymphocytes in the diet supplemented with 33 mg/kg flavanone significantly increased the ratio of cells in S phase and G 2 / M phase (P 0.05N 3). The apoptosis rate of peripheral blood lymphocytes in the control group and 33 mg/kg group was significantly higher than that in the control group and 11 mg/kg group.It can be seen that the addition of 1133 mg/kg flavanone in diet can promote the transformation of peripheral blood lymphocytes and the transformation of cells to DNA synthesis, promote cell division, and promote apoptosis.The best effect was obtained by adding 33 mg/kg flavonoids from Allium mongolicum.
