湖南濱湖水牛卵泡動態(tài)發(fā)育及生殖激素變化的研究

【摘要】:為了提高濱湖水牛繁殖力,本研究運用同期發(fā)情技術(shù)和B超診斷分析了濱湖水牛卵泡發(fā)育動態(tài)變化和生殖激素含量變化。1 湖南濱湖水牛卵泡發(fā)育動態(tài)變化的研究本研究采用同期發(fā)情技術(shù)處理濱湖水牛,利用B超檢測濱湖水牛卵泡動態(tài)發(fā)育情況。研究結(jié)果表明:(1)卵泡波出現(xiàn)時間:2波周期青年水牛的第1波和第2波分別在1.5d,8.2d出現(xiàn);經(jīng)產(chǎn)水牛則在1.0d,12.3d出現(xiàn),兩者差異不顯著(P0.05)。3波周期中第1波、第2波、第3波分別出現(xiàn)在1.0d,7.5d和15.0d。(2)卵泡波持續(xù)時間:2波周期中,青年牛第1波、第2波分別持續(xù)7.16d、10.33d;經(jīng)產(chǎn)牛第1波、第2波分別為9.33d和11.13d。3波周期中3個波各持續(xù)6.5、8.0和9.0d。2波或3波周期,各波持續(xù)的天數(shù)間均無顯著性差異(P0.05)。(3)發(fā)情周期內(nèi)征集的卵泡總數(shù):經(jīng)產(chǎn)牛為16.46個,要顯著高于青年牛的12.48個(P0.05)。(4)周期內(nèi)選擇的卵泡總數(shù):經(jīng)產(chǎn)牛為7.96個,與青年牛的7.30個差異不顯著(P0.05)。(5)優(yōu)勢卵泡最大直徑:2波周期中青年牛第1、2波優(yōu)勢卵泡最大直徑分別為8.22mm和9.81mm,經(jīng)產(chǎn)牛第1、2波優(yōu)勢卵泡最大直徑分別為7.85mm和7.54mm,兩者差異不顯著(P0.05)。3波周期中的濱湖青年母水牛的第1、2、3波優(yōu)勢卵泡最大直徑分別為7.54mm、10.33mm和8.32mm。(6)各級卵泡數(shù):青年水牛、經(jīng)產(chǎn)水牛之間在左、右卵巢上各類型卵泡的數(shù)量統(tǒng)計均無明顯差異(P0.05)。2濱湖水牛生殖激素變化的研究本研究運用放射免疫技術(shù)分析了發(fā)情周期內(nèi)生殖激素變化。研究結(jié)果表明:(1)從發(fā)情前2d到出現(xiàn)發(fā)情,FSH持續(xù)升高,發(fā)情后1d出現(xiàn)最高峰(3.5mIU/mL),然后持續(xù)回落。按發(fā)情周期各階段分析,發(fā)情中期FSH水平值最高,且顯著高于發(fā)情前期、發(fā)情后期和間情期(P0.05);(2)從發(fā)情前期到發(fā)情第3d,LH水平呈現(xiàn)一個緩慢上升的過程,其中第3d達(dá)到22.13mIU/mL,第6d降到16.12mIU/mL;然后先緩慢升高再下降,第10d達(dá)到最低值15.17mIU/mL;之后緩慢升高,分別在14d、16d出現(xiàn)兩個小高峰,峰值分別為20.71和22.17mIU/mL,第18d出現(xiàn)最高值23.24mIU/mL。發(fā)情期三個階段(前期、中期和后期)的LH水平分別為20.65、20.02和21.29mIU/mL,但差異不顯著(P0.05),但都要顯著高于間情期的17.24mIU/mL(Pp0.05)。(3)發(fā)情前2d,E2水平最低(20.43 pg/mL),然后持續(xù)緩慢升高,發(fā)情第6d達(dá)到一個小高峰(72.88 pg/mL);之后緩慢下降,第15d到達(dá)低值(39.90 pg/mL);之后迅速上升,第19d達(dá)到最大峰值(118.32 pg/mL),隨后又迅速下降到40pg/mL以下。發(fā)情前期E2水平(67.83 pg/mL)顯著高于發(fā)清中期(51.01 pg/mL)、發(fā)情后期(56.33pg/mL)和間情期(53.74 pg/mL) (P0.05)。而其他各階段的E2水平差異不顯著(Pp0.05)。(4)發(fā)情前2d,P_4水平急劇下降,發(fā)情當(dāng)天(0d)降到最低值(0.37pg/mL),然后開始上升(出現(xiàn)三個峰值,即第7d為6.79pg/mL,10d達(dá)到最大值8.96pg/mL,14d為7.77pg/mL);隨后迅速下降。發(fā)情中期(1.87pg/mL)和發(fā)情后期(2.62pg/mL) P_4水平最低,兩者差異不顯著和(P0.05),但均低于發(fā)情前期(4.13pg/mL和間情期(5.71pg/mL),且差異顯著(P0.05),間情期P_4水平高于發(fā)情前期,且差異顯著(P0.05)。

The results show that: (1) from 2D to appear before estrus estrus, FSH continues to rise, the peak estrus after 1D (3.5mIU/mL), and then continued to fall.
