欽州地區(qū)禽白血病的流行調(diào)查及其分離株env基因的分析

【摘要】:禽白血病(Avian leukosis, AL)是由統(tǒng)稱為禽白血病病毒(Avian leukosis virus, ALV)的一群病毒引起的多種腫瘤病的總稱。ALV給養(yǎng)禽業(yè)造成巨大的經(jīng)濟(jì)損失,國內(nèi)外都給予很大關(guān)注,目前在全國范圍內(nèi)已經(jīng)開展流行病學(xué)調(diào)查和凈化工作。廣西自開展AL的研究和凈化工作以來,對玉林、桂林、百色、南寧等主要養(yǎng)禽地區(qū)的流調(diào)工作已基本完成。本課題選擇廣西的臨海地區(qū)——?dú)J州進(jìn)行相關(guān)的流行病學(xué)調(diào)查以及病毒的分離和基因序列測定與分析,旨在完善全區(qū)范圍內(nèi)的主要地方品種雞及其周圍飼養(yǎng)的水禽及其他禽類的流行病學(xué)資料,并通過病毒分離和對分離株重要基因進(jìn)行測序分析與比較,進(jìn)一步掌握當(dāng)?shù)厍蓊惛腥続LV的亞型及分離株的遺傳變異情況,為當(dāng)?shù)氐膬艋c防控工作提供依據(jù)。首先,課題采集欽州地區(qū)飼養(yǎng)的鐵腳麻雞、“海鴨蛋”麻鴨、獅頭鵝、鴿子和火雞5個主要家禽品種的肛拭、蛋清和血清共計953份樣品,應(yīng)用IDEXX生產(chǎn)的商品化ALV-p27抗原和A/B、J抗體ELISA檢測試劑盒進(jìn)行檢測。結(jié)果顯示,除了鐵腳麻雞的肛拭樣品之p27抗原陽性率為13.04%(12/92)、蛋清樣品之p27抗原陽性率為3.26%(3/92)及血清樣品之A/B抗體陽性率為5.08%(3/59)之外,其它樣品及項目的檢測的陽性率均為0%。其次,課題對抗原或抗體檢測為陽性的16份樣品、鐵腳麻雞檢測陰性的16份樣品以及火雞檢測陰性的33份樣品共65份樣品相對應(yīng)的禽只的血清進(jìn)行DF-1細(xì)胞培養(yǎng)的病毒分離,再用ELISA和PCR的方法分別檢測其F2代的細(xì)胞培養(yǎng)液和培養(yǎng)物,經(jīng)鑒定為陽性的樣品再進(jìn)行病毒e7nv基因的擴(kuò)增及其序列的測定與分析。結(jié)果從65份樣品中分離出2株J亞群ALV(分別命名為QZ2和QZ14),QZ2和QZ14的env基因核苷酸同源性很高(95.7%);它們gp37基因核苷酸及其編碼的氨基酸與J亞群參考株的同源性都在90%以上,env與gp85基因核苷酸與J亞群參考株(除ADOL-7501外)的同源性也都在90%以上,而gp85基因編碼的氨基酸與J亞群參考株的同源性在70.5%~90.9%之間,與原型株HPRS103的達(dá)90%以上,U3區(qū)基因核苷酸與J亞群參考株的同源性在87.6%~92.9%之間,QZ2的E元件基本保持完整,而QZ14有5個堿基的缺失。本課題研究的結(jié)果表明,欽州地區(qū)5個主要禽類中僅鐵腳麻雞存在ALV的感染,經(jīng)分離鑒定為J亞群,基因序列分析表明QZ2和QZ14的不同基因已發(fā)生變異。
[Abstract]:Avian leukemia (ALV) is caused by a group of viruses called avian leukemia virus (ALV). ALV has caused enormous economic losses to poultry industry, and has attracted great attention both at home and abroad.At present, epidemiological investigation and purification have been carried out nationwide.Since the research and purification of AL in Guangxi, the flow regulation of the main poultry breeding areas such as Yulin, Guilin, Baise and Nanning has been basically completed.In this study, we selected Linhai region of Guangxi, Qinzhou to carry on the related epidemiology investigation, the virus isolation and the gene sequence determination and analysis.The aim of this study was to improve the epidemiological data of major local chickens and their surrounding waterfowl and other birds in the whole region, and to analyze and compare the important genes of the isolates by virus isolation and sequencing.To further understand the subtypes of ALV infection and the genetic variation of isolates in local poultry, to provide the basis for local purification, prevention and control.First of all, 953 samples of egg white and serum were collected from five main poultry breeds in Qinzhou, namely, hens, sea duck, lion goose, pigeon and turkey.The commercial ALV-p27 antigen produced by IDEXX and the ELISA kit of A / B J antibody were used for detection.The results showed that the positive rate of p27 antigen in anus swabs was 13.04 / 92%, that of egg white was 3.26 / 92) and the positive rate of A / B antibody in serum was 5.08 / 3 / 59), the positive rates of other samples and items were all 0% (P < 0.05), the results showed that the positive rate of p27 antigen in anus swab samples was 13.04% / 92%, 3.26% / 92% in egg white samples, and 5.08% / 59 in serum samples respectively.Secondly, 16 samples with positive antigens or antibodies, 16 samples with negative detection of iron-footed hens and 33 samples with negative turkey were isolated from sera of poultry corresponding to DF-1 cell culture.ELISA and PCR were used to detect the culture medium and culture of F2 passage respectively. The positive samples were amplified by e7nv gene and sequenced.Results two strains of subgroup J ALV (named QZ2 and QZ14, QZ2 and QZ14) were isolated from 65 samples. The nucleotide homology of gp37 gene and its encoded amino acid was 90%.The nucleotides of gp85 gene and the reference strain of subgroup J (except ADOL-7501) were all more than 90% homology.The homology between amino acids encoded by gp85 gene and reference strain of subgroup J was between 70.5% and 90.9%, and the homology of E element of QZ2 between gp85 gene and the reference strain of subgroup J was 87.6% and 92.9%, respectively.QZ14 has a 5-base deletion.The results of this study showed that ALV was isolated and identified as subgroup J among the five main poultry species in Qinzhou area. The gene sequence analysis showed that the different genes of QZ2 and QZ14 had been mutated.
