靜原雞ELOVL2和ELOVL5基因表達(dá)的組織特異性研究

【摘要】:采用實時熒光定量PCR(qRT-PCR)技術(shù)對寧夏地方優(yōu)良品種靜原雞的肝臟、肌肉、心臟、睪丸、卵巢、腹脂6個組織中ELOVL2和ELOVL5基因的表達(dá)量進行了測定,并通過SAS8.2軟件進行分析統(tǒng)計。結(jié)果表明,ELOVL2和ELOVL5基因在靜原雞6個不同組織中均有表達(dá)。其中ELOVL2基因在肝臟中的相對表達(dá)量最高,為71.52±0.41,顯著高于其他組織(P0.05);其次是卵巢、睪丸、腹脂、心臟及肌肉,在另外5個組織中表達(dá)量雖有差異,但均差異不顯著(P0.05)。ELOVL5基因在靜原雞不同組織中的相對表達(dá)量依次為:肝臟腹脂卵巢睪丸心臟肌肉,同樣在肝臟中的表達(dá)量最高,為110.94±0.02,顯著高于其他組織(P0.05),在腹脂中的表達(dá)量顯著高于卵巢、睪丸、心臟和肌肉(P0.05)。另外在心臟和肌肉中的表達(dá)量均差異不顯著(P0.05)。對ELOVL2和ELOVL5基因分別在不同組織中的表達(dá)差異進行分析比較,發(fā)現(xiàn)兩個基因在腹脂中的表達(dá)量差異顯著(P0.05),ELOVL5基因在腹脂中的表達(dá)量較高。研究結(jié)果說明,ELOVL2和ELOVL5基因在靜原雞6種組織中的表達(dá)存在差異,并以肝臟組織最為明顯,這為地方品種的開發(fā)與利用、種質(zhì)資源遺傳評定及地方品種分子育種技術(shù)平臺的構(gòu)建奠定了一定的理論基礎(chǔ)。
[Abstract]:The results showed that both ELOVL2 and ELOVL5 genes were expressed in 6 different tissues of Chicken.The relative expression of ELOVL2 gene in liver was the highest (71.52 鹵0.41), which was significantly higher than that in other tissues (P 0.05), followed by ovary, testis, abdominal fat, heart and muscle.However, the relative expression of P0.05N. ELOVL5 gene in different tissues of Chicken was in the order of liver, abdominal fat, ovary, testis, heart muscle, and the highest expression in liver.The expression of P0.05 in abdominal fat was significantly higher than that in ovary, testis, heart and muscle.In addition, there was no significant difference in the expression of P0.05 in the heart and muscle.The differences of expression of ELOVL2 and ELOVL5 genes in different tissues were analyzed and compared. It was found that the expression of ELOVL2 and ELOVL5 genes in abdominal fat was significantly different, and the expression of ELOVL5 gene in abdominal fat was higher than that in abdominal fat.The results showed that there were differences between EloVL2 and ELOVL5 gene expression in six tissues of Jingyuan chicken, and the liver tissue was the most obvious, which was the development and utilization of local breeds.Genetic evaluation of germplasm resources and the construction of molecular breeding platform for local varieties lay a certain theoretical foundation.
