各型FMDV VP1基因表位嵌入型蛋白SLP-MultiVP1對免疫不同小鼠血清IgG亞型的影響

【摘要】:利用通過Swiss-Model在線軟件分析所得的乳桿菌S-層蛋白高級結構3D模擬圖,應用Swiss-Pbd Viewer軟件對嵌入型蛋白SLP-MultiVP1中的3型口蹄疫病毒(FMDV)不同表位位點進行標注,并用嵌入型蛋白SLP-MultiVP1和重組蛋白SLP分別免疫6周齡的BALB/c和昆明小鼠,分離3次免疫后的血清,用Mouse IgG1Ready-SET-Go和Mouse IgG2aReady-SET-Go ELISA試劑盒檢測血清中IgG抗體亞型IgG1和IgG2a。通過SPSS統(tǒng)計學分析發(fā)現:嵌入型蛋白SLP-MultiVP1組IgG1和IgG2a水平顯著高于SLP組(P0.05),其中嵌入型蛋白SLP-MultiVP1免疫組的IgG2a顯著高于IgG1(P0.05),但是BALB/c和昆明小鼠之間無顯著性差異(P0.05)。該結果提示,嵌入型蛋白SLP-MultiVP1可能傾向于激發(fā)Th1類細胞介導的免疫應答,而且無個體差異。
[Abstract]:Using 3D simulation map of Lactobacillus Slayer protein advanced structure obtained by Swiss-Model online software, different epitopes of FMDV3 in embedded protein SLP-MultiVP1 were labeled by Swiss-Pbd Viewer software. The 6-week-old BALB/c and Kunming mice were immunized with embedded protein SLP-MultiVP1 and recombinant protein SLP respectively. Mouse IgG1Ready-SET-Go and Mouse IgG2aReady-SET-Go ELISA kit were used to detect IgG antibody subtype IgG1 and IgG2a in serum. SPSS statistical analysis showed that IgG1 and IgG2a levels in SLP-MultiVP1 group were significantly higher than those in SLP group, and IgG2a in SLP-MultiVP1 immunized group was significantly higher than that in SLP group. The results suggest that there is no significant difference between BALB/c and Kunming mice. The embedded protein SLP-MultiVP1 may be inclined to stimulate the immune response mediated by Th1 cells, and there is no individual difference.
