3~6月齡伊犁馬腸道微生物群落多樣性的研究

【摘要】:本試驗(yàn)旨在研究3~6月齡伊犁馬腸道微生物群落的多樣性,揭示斷奶前馬駒腸道菌群演替的規(guī)律,從微生物學(xué)角度為此階段馬駒生長發(fā)育及腸道健康研究提供理論依據(jù)。試驗(yàn)選取平均體重為(89.75±8.81)kg、出生日期相同的3月齡伊犁馬5匹,進(jìn)行為期90 d的飼養(yǎng)試驗(yàn)。分別在試驗(yàn)的第0天(即試驗(yàn)開始的前1天)、第30天、第60天、第90天采集馬駒糞便樣品,從每份樣品中提取微生物基因組總DNA,采用Illumina HiSeq測序技術(shù)檢測樣品微生物群落多樣性。結(jié)果顯示:1)對5匹馬駒的20份糞樣進(jìn)行測序,共獲得有效序列數(shù)157 665條,獲得平均操作分類單元(OTUs)1 117個。2)馬駒糞便中微生物群落α多樣性指數(shù)(ACE、Chao1、Shannon和Simpson指數(shù))隨著馬駒月齡的增加呈波動性變化,但各時間點(diǎn)之間均無顯著性變化(P0.05)。3)在門水平上,馬駒糞便中10大優(yōu)勢菌分別為厚壁菌門(Firmicutes)、擬桿菌門(Bacteroidetes)、變形菌門(Proteobacteria)、疣微菌門(Verrucomicrobia)、螺旋體菌門(Spirochaetes)、梭桿菌門(Fusobacteria)、無壁菌門(Tenericutes)、放線菌門(Actinobacteria)、TM7、廣古菌門(Euryarchaeota),其中以厚壁菌門、擬桿菌門、變形菌門及疣微菌門的豐度較高;在科水平上,馬駒糞便中10大優(yōu)勢菌分別為芽孢桿菌科(Bacillaceae)、莫拉氏菌科(Moraxellaceae)、動球菌科(Planococcaceae)、肉桿菌科(Carnobacteriaceae)、BS11、RFP12、乳桿菌科(Lactobacillaceae)、瘤胃球菌科(Ruminococcaceae)、毛螺旋菌科(Lachnospiraceae)、紫單胞菌科(Porphyromonadaceae);在屬水平上,馬駒糞便中10大優(yōu)勢菌分別為不動桿菌屬(Acinetobacter)、德庫菌屬(Desemzia)、乳桿菌屬(Lactobacillus)、Ureibacillus、Paludibacter、芽孢桿菌屬(Bacillus)、埃希式桿菌屬(Escherichia)、肉食桿菌屬(Carnobacterium)、密螺旋體屬(Treponema)、艱難桿菌屬(Mogibacterium)。由此得出,采用Illumina HiSeq測序技術(shù)能夠準(zhǔn)確地對3~6月齡伊犁馬腸道微生物群落進(jìn)行分類研究;厚壁菌門、擬桿菌門、變形菌門、疣微菌門是3~6月齡伊犁馬腸道內(nèi)的優(yōu)勢菌門。
[Abstract]:The aim of this study was to study the diversity of intestinal microflora in the yili horse at the age of 3 and 6 months, and to reveal the succession of intestinal microflora before weaning. From the point of view of microbiology, the theoretical basis was provided for the study of growth, development and intestinal health of the foal at this stage. The average body weight was 89.75 鹵8.81 kg, and 5 horses of the same birth date were 3 months old Yili horses. A 90-day feeding trial was conducted. Samples of foal dung were collected on day 0 (1 day before the trial, 30 days, 60 days, 90 days), respectively. The total genomic DNA of microorganism was extracted from each sample, and the diversity of microbial community was detected by Illumina HiSeq sequencing technique. The results showed that 20 feces from 5 ponies were sequenced, and the effective sequence number was 157,665. The average operating classification unit (OTUs1 117. 2) the 偽 -diversity index of microbial community in the feces of pony (ACE-Chao1 Shannon and Simpson index) fluctuated with the increase of foal age, but there was no significant change at each time point (P 0.05N. 3) at the gate level. The top 10 dominant bacteria in the feces of pony are Firmicuteschus, Bacteroidetesa, Proteobacteriae, Verrucomicrobiae, Spirochaetesa, Fusobacteria, Tenericuteschus, Actinobacteriae TM7, Euryarchaetataa. The abundance of the phylum Proteus and Microphylum verruca is higher; at the family level, The top 10 dominant bacteria in the feces of pony are Bacillaceaeae, Moraxellaceaeae, Planoacecoccae, Carnobacter aceaeus BS11RFP12, Lactobacillus aceaeae, Ruminococcaceaeae, Lachnospiraceaeae, Porphyromonadaceaeae; at the generic level, The dominant bacteria in the feces of pony are Acinetobacter Acinetobacter, Desemziaus, Lactobacillus Ureibacillus Paludibacterium, Bacillus, Escherichia coli, Carnobacterium, Treponemia, Mogibacterium iumum. Using Illumina HiSeq sequencing technique, the intestinal microflora of Yili horse at the age of 3 and 6 months could be accurately classified and studied, and the phylum platyphylum, Bacteroides phylum, Proteus phylum and Microphylum verrucum were the dominant microphylum in the intestinal tract of Yili horse at the age of 3 or 6 months.
