免疫共沉淀聯(lián)合質(zhì)譜技術(shù)篩選宿主因子CypB的互作蛋白

【摘要】:為探討親環(huán)蛋白B(cyclophilin B,CypB)促進(jìn)羊傳染性膿皰病毒(orf virus,ORFV)復(fù)制增殖的機(jī)制,以O(shè)RFV早期感染MDBK細(xì)胞為試驗材料,利用免疫共沉淀技術(shù)篩選與CypB互作的蛋白。免疫共沉淀產(chǎn)物的Western blot和SDS-PAGE的分析結(jié)果顯示,在大小約為25 000處可見有明顯的差異條帶。切下該差異蛋白條帶進(jìn)行質(zhì)譜鑒定分析,將獲得的原始質(zhì)譜數(shù)據(jù)使用Mascot軟件中的串級質(zhì)譜數(shù)據(jù)搜索功能(MS/MS Ions Search)進(jìn)行搜索鑒定,初步篩選獲得6個可能與CypB相互作用的病毒相關(guān)蛋白。為進(jìn)一步深入研究CypB在ORFV早期侵染過程中的作用奠定基礎(chǔ)。
[Abstract]:In order to investigate the mechanism of cyclophilin BypB) in promoting the replication and proliferation of sheep infectious pustules virus (ORFV), the early infection of MDBK cells with ORFV was used as the experimental material. The immunoprecipitation technique was used to screen proteins interacting with CypB. The results of Western blot and SDS-PAGE analysis of the immunoprecipitation products showed that there were obvious differential bands at about 25 000. The differential protein bands were cut off and identified by mass spectrometry. The original mass data will be searched and identified using the Cascade MS Ions search function in the Mascot software. Six virion related proteins which may interact with CypB were obtained, which laid a foundation for further study on the role of CypB in the early infection of ORFV.
